Depressed Symptomatology in Systemic Lupus Erythematosus Patients

Summary: This study examined the longitudinal course of depressed affect among outpatients with systemic lupus erythematosus (SLE) and the relationship between SLE disease activity and depressed affect. The majority of patients had persistent depressed affect or major depression over a period of up to 4 years, and there was no consistent relationship between depressed affect and SLE disease activity.

The Role of Complement in Autoimmune Disease-Associated Thrombotic Microangiopathy and the Potential for Therapeutics

Summary: The complement system is a crucial component connecting the innate and humoral immune systems, but it can also be damaging to host tissue if misdirected or dysregulated. It plays an important role in atypical hemolytic uremic syndrome (aHUS), a thrombotic microangiopathy characterized by microangiopathic hemolytic anemia, thrombocytopenia, and organ injury. However, the role of complement in TMA associated with other diseases and its interaction with neutrophils and coagulation pathways complicates the understanding of its involvement. Understanding the role of complement in TMA associated with autoimmune diseases will help in timely diagnosis and management.

Summary: The Renal Activity Index for Lupus (RAIL) can non-invasively identify lupus nephritis (LN) and assess the efficacy of LN induction therapy. RAIL scores can differentiate clinically active LN from inactive LN or without LN. RAIL scores significantly decrease after complete LN remission.

Systematic review of colchicine neuromyopathy: Risk factors, duration and resolution

Summary: This study aimed to identify reports of colchicine-induced neuropathy and myopathy and determine the risk factors associated with this toxicity at commonly used doses. A systematic review of case reports found 143 cases of neuromyopathy associated with the use of colchicine. Most cases presented with features of both neuropathy and myopathy, with symptoms of myopathy being predominant. Cessation of colchicine generally led to complete resolution of symptoms.

Summary: This study assessed the impact of the COVID-19 pandemic on trainees and early career investigators and proposed ways for the ACR and RRF to address their needs. The survey revealed that 75% of respondents pursuing academic research careers had to put their primary projects on hold during the pandemic. The suggested interventions included increased funding, funding process reform, and expanding mentoring and networking resources.

Improving the sensitivity of in vivo CRISPR off-target detection with DISCOVER-Seq

Summary: DISCOVER-Seq+ is a highly sensitive method that detects off-target effects of CRISPR genome editing. By inhibiting DNA-dependent protein kinase catalytic subunit and accumulating the repair protein MRE11 at CRISPR-Cas-targeted sites, off-target sites can be mapped to positions of MRE11 binding using chromatin immunoprecipitation followed by sequencing. This technique, DISCOVER-Seq+, has discovered up to fivefold more off-target sites compared to previous methods in various cell lines and animal models.

Summary: This study tested samples from immunocompetent adults and those receiving treatments for chronic inflammatory diseases before and after mRNA SARS-CoV-2 vaccination. The lateral flow assay (LFA) detected anti-S antibodies in the majority of participants and showed significant correlation with ELISA results. LFA could be a practical alternative for detecting antibodies after infection or vaccination, particularly in outpatient or resource-limited settings.

Making the case for precision dosing: visualizing the variability of cefepime exposures in critically ill adults

Summary: A study was conducted to investigate the variability in cefepime exposures among real-world ICU patients using modeling and simulations. The results showed wide variations in cefepime exposure among patients with similar kidney function estimates. Current population adjustment schemes based solely on creatinine clearance may result in unintended high and low drug exposures, raising concerns about safety and efficacy.

Summary: This study investigated factors associated with severe COVID-19 in patients with psoriasis, psoriatic arthritis, and axial spondyloarthritis. The study found that age, sex, comorbidities, disease activity, and glucocorticoid use were associated with the severity of COVID-19. Additionally, later pandemic time periods, psoriasis, and the use of TNFi, IL17i, and IL-23i/IL-12+23i were associated with a reduced risk of severe COVID-19.
