Structural studies of the rhamnose-rich cell wall polysaccharide of Lactobacillus casei BL23

Lactobacillus casei is a Gram positive lactic acid bacterium used in dairy fermentations and present in the normal human gut microbiota. Certain strains are recognized as probiotics with beneficial effects on human and animal health. L. casei BL23 is a potential probiotic strain endowed with anti-inflammatory properties and a model strain widely used in genetic, physiological and biochemical studies. A number of bacterial cell surface polysaccharides have been shown to play a role in the immune modulation activities observed for probiotic lactic acid bacteria. In the present work, we purified the most abundant carbohydrate polymer of L. casei BL23 cell wall, a neutral wall polysaccharide (WPS) and established its chemical structure by periodate oxidation, methylation analysis and 2D NMR spectroscopy. The WPS of L. casei BL23 was shown to contain alpha-Rha, alpha-Glc, beta-G1cNAc and beta-GaINAc forming a branched heptasaccharide repeating unit (variant 1) with an additional partial substitution with alpha-Glc (variant 2). A modified non-reducing end octasaccharide, corresponding to a terminal unit of the WPS (variant 3), was also identified and allowed to define the biological repeating unit of the WPS. To our knowledge, this is the first report of the identification of a biological repeating unit based on a chemical evidence, in a cell wall polysaccharide of a Gram positive bacterial species. Characterization of the marine aquaculture microbiome: A seasonal survey in a seabass farm

Summary: This study conducted in a seabass farm in northern France revealed that bacterial communities in biofilms and tanks were more diverse compared to water inlets. The majority of phylum diversity was composed of Proteobacteria and Bacteroidetes, with Vibrionaceae family isolates predominantly found in tank water samples. The persistence of Vibrionaceae in biofilms suggests their potential role as a reservoir in case of dysbiosis in aquaculture environments.

Pili and other surface proteins influence the structure and the nanomechanical properties of Lactococcus lactis biofilms

Summary: Lactic acid bacteria, especially Lactococcus lactis, are widely used in the food industry for the control and protection of fermented food manufacturing processes. Recent studies have shown that certain strains of lactic acid bacteria form complex biofilms with heterogeneous and aerial structures. The presence of pili in these strains affects the biomechanical properties of the biofilms, with non-piliated strains being smoother and stiffer compared to piliated strains. Additionally, the role of pili in biofilm cohesiveness was demonstrated through the adhesion of bacteria to the biofilm surface.

Summary: The study highlights the threat of vibriosis in seabass aquaculture, mainly caused by Vibrio harveyi. Monitoring the presence of V. harveyi in water and biofilms over time is crucial for understanding the dynamics of the pathogen. The constant presence of V. harveyi in rearing tanks and water inlets suggests an increased risk of vibriosis outbreaks, especially with rising water temperatures.

Summary: The human gut symbiont Lacticaseibacillus (L.) casei has a wide range of immunomodulatory effects on the human host. This study demonstrates that human monocyte-derived dendritic cells (moDCs) can engulf L. casei BL23, and the intact bacterial cell wall and morphology play a key role in this process. The absence of the bacterial cell-wall-degrading enzyme Lc-p75 leads to defective internalization and impaired cytokine secretion by moDCs, resulting in reduced T-cell activation capacity. Formation of short bacterial chains helps to exert the potent immunomodulatory properties of L. casei BL23.

Summary: In this study, the cytoplasmic steps of the TA d-alanylation pathway in Lactiplantibacillus plantarum were investigated using structural biology and in vivo approaches. The protein interactions involved in this pathway were characterized, with DltC1 identified as a central hub for the cytoplasmic steps. The importance of AcpS, DltB, DltC1, and DltA in promoting Drosophila juvenile growth under chronic undernutrition was also established.

The D-Ala-LTAs are shown to be important cues supporting intestinal peptidase expression and juvenile growth in Drosophila melanogaster.
