Summary: Oral administration is a convenient way to treat gastrointestinal diseases with good patient compliance, but the nonspecific distribution of drugs can lead to serious side effects. To overcome this, oral drug delivery systems (ODDS) have been developed, but they are limited by physiological barriers in the gastrointestinal tract. Micro/nanomotors (MNMs) have shown potential in targeted drug delivery, and this review comprehensively discusses their applications in overcoming physiological barriers in ODDS for gastrointestinal diseases. This review provides inspiration and direction for MNMs in the therapy of gastrointestinal diseases and advances the clinical application of MNMs in oral drug delivery.

However, the complexity of the biological environment and the particularity of disease microenvironment hinder their advances. This review comprehensively addresses the design, imaging, and disease treatment of biomedical MNMs, highlighting intelligent MNMs with biomimetic functionality and chemotactic capability, emphasizing the applicability of different imaging techniques, and focusing on various proof-of-concept studies for the treatment of major diseases. The challenges and limitations of current biomedical MNMs are discussed, inspiring future research towards clinical translation.

Summary: This study describes an effective strategy for delivering intracellular antigens using ultrasound-propelled gold nanowires nanomotors modified with a model antigen. The nanomotors facilitate rapid internalization into antigen-presenting cells, leading to activation and cross-presentation of the cells with significant increase in MHC I and MHC II molecules expression levels. This novel vaccine delivery strategy based on AuNWs nanomotors is expected to enhance cellular immunity and provide a new approach to immunotherapy.
