Summary: B7-H3 is highly expressed in various types of cancer and plays important roles in tumor immune evasion, carcinogenesis, metastasis, and angiogenesis. This study investigated the effects of cancer cell-derived exosomal B7-H3 on tumor angiogenesis and metastasis. The findings showed that exosomal B7-H3 from colorectal cancer cells was taken up by endothelial cells and activated the AKT1/mTOR/VEGFA signaling pathway, enhancing their migration, invasion, and tube formation abilities. Furthermore, exosomes with reinforced B7-H3 promoted pulmonary angiogenesis and metastasis in mice. High expression of B7-H3 was also observed in urinary exosomes from colorectal cancer patients.

Summary: This study explored the functional mechanism of action of LINC00460 in cervical cancer angiogenesis for the first time. The results showed that conditioned medium from LINC00460-knockdown cervical cancer cells attenuated human umbilical vein endothelial cell (HUVEC) migration, invasion, and tube formation, while LINC00460 upregulation had the opposite effects. Mechanistically, LINC00460 stimulated VEGFA transcription. The study suggests that the NF-KB-VEGFA axis activated by LINC00460 is a promising target for blocking tumor angiogenesis.

Summary: In the context of obesity, inflammatory cytokines secreted by adipocytes contribute to insulin resistance and play a key role in the development of metabolic syndrome. Our previous study found that the transcription factor KLF7 promotes the expression of p-p65 and IL-6 in adipocytes, but the specific molecular mechanism remains unclear. In this study, we found that the expression of KLF7, PKC?, p-I?B, p-p65, and IL-6 was significantly increased in epididymal white adipose tissue (Epi WAT) of mice fed a high-fat diet (HFD). However, in KLF7 fat conditional knockout mice, the expression of PKC?, p-I?B, p-p65, and IL-6 was significantly decreased in Epi WAT. In 3T3-L1 adipocytes, KLF7 promotes the expression of IL-6 through the PKC?/NF-?B pathway. Furthermore, luciferase reporter and chromatin immunoprecipitation assays confirmed that KLF7 upregulates the expression of PKC? transcripts in HEK-293T cells. Together, our results demonstrate that KLF7 promotes the expression of IL-6 by upregulating PKC? expression and activating the NF-?B signaling pathway in adipocytes.
