mTOR/HDAC1 Crosstalk Mediated Suppression of ADH1A and ALDH2 Links Alcohol Metabolism to Hepatocellular Carcinoma Onset and Progression in silico

Hepatocellular carcinoma (HCC) is ranked the third deadliest cancer worldwide whose molecular pathogenesis is not fully understood. Although deregulated metabolic pathways have been implicated in HCC onset and progression, the mechanisms triggering this metabolic imbalance are yet to be explored. Here, we identified a gene signature coding catabolic enzymes (Cat-GS) involved in key metabolic pathways like amino acid, lipid, carbohydrate, drug, and retinol metabolism as suppressed in HCC. A higher expression of deregulated Cat-GS is associated with good survival and less aggressive disease state in HCC patients. On the other hand, we identified mTOR signaling as a key determinant in HCC onset and progression, whose hyperactivation is found associated with poor survival and aggressive disease state in HCC patients. Next, out of Cat-GS, we established two key regulators of alcohol metabolism, alcohol dehydrogenase 1A (ADH1A) and aldehyde dehydrogenase 2 (ALDH2), as being transcriptionally suppressed by histone deacetylase 1 (HDAC1) at the downstream of mTORC1 signaling. Suppressed ADH1A and ALDH2 expression aligns well with HCC-specific molecular profile and can efficiently predict disease onset and progression, whereas higher ADH1A and ALDH2 expression is associated with good survival and less aggressive disease state in HCC patients. Overall, our in silico findings suggest that transcriptional suppression of alcohol metabolism regulators, ADH1A and ALDH2, at the downstream of mTOR signaling is, in part, responsible for triggering oncogenic transformation of hepatocytes resulting in disease onset and progression in HCC.

Targeting HIF1-alpha/miR-326/ITGA5 axis potentiates chemotherapy response in triple-negative breast cancer

Summary: In this study, we identified fibronectin (FN1) as a central driver gene in chemoresistance. Overexpression of miR-326 reversed FN1-driven chemoresistance by targeting the FN1 receptor, ITGA5. miR-326 expression was downregulated by increased hypoxia/HIF1A and ECM stiffness in chemoresistant tumors, leading to upregulation of ITGA5 and activation of the downstream FAK/Src signaling pathways. Overexpression of miR-326 or inhibition of ITGA5 overcame FN1-driven chemotherapy resistance in vitro by inhibiting the FAK/Src pathway and potentiated the efficacy of chemotherapy in vivo. Importantly, lower expression of miR-326 or higher levels of predicted miR-326 target genes were significantly associated with worse overall survival in chemotherapy-treated TNBC patients.

Alpha-1-antichymotrypsin as a novel biomarker for diagnosis, prognosis, and therapy prediction in human diseases

Summary: This review summarizes the expression level, glycosylation modification, and biological characteristics of the glycoprotein AACT in inflammation, neurodegenerative and elderly diseases, and tumorigenesis. AACT's dysregulation and glycosylation levels are associated with tumor progression and recurrence, making it a potential biomarker for tumor monitoring. Additionally, AACT's aberrant expression in cancer may be due to genetic alterations and/or immune regulation, and it may serve as a diagnostic or prognostic biomarker or therapeutic target in tumors. The review confirms the key roles of AACT in tumorigenesis, suggesting its potential importance as a biomarker for tumor diagnosis and prognosis, as well as a therapeutic target for human diseases.

Decoding ceRNA regulatory network in the pulmonary artery of hypoxia-induced pulmonary hypertension (HPH) rat model

Summary: A ceRNA regulatory network was constructed in the pulmonary arteries of HPH rats, identifying 10 hub miRNAs and their corresponding interacting lncRNAs, circRNAs, and mRNAs. The expression patterns of selected DERNAs were validated to be consistent with the sequencing result. POSTN, LTBP2, SPP1, and LSAMP were suggested as potential diagnostic biomarkers and therapeutic targets for PAH.

A unified global tractography framework for automatic visual pathway reconstruction

Progressive structural and covariance connectivity abnormalities in patients with Alzheimer's disease

Summary: This study investigates the grey matter atrophy, structural covariance connectivity abnormalities, and the correlations between grey matter atrophy and cognitive decline in Alzheimer's disease (AD) patients. The results show significant grey matter atrophy in multiple brain regions of AD patients, and the decrease in grey matter volume is positively correlated with cognitive decline. Additionally, abnormal structural covariance connectivity is observed in AD patients.

2-Hydroxypropyl Group Linked Derivatives of Indole Azoles as Potential Multifunctional Antibacterial Candidates for Effectively Inhibiting the Activity of MRSA and Responding Inflammatory Factors

Summary: The development of new structural agents with significant antimicrobial potential is necessary due to the increasing resistance of methicillin-resistant Staphylococcus aureus (MRSA) to antibiotics. A novel class of 2-hydroxypropyl group linked derivatives of indole azoles was found to have excellent antibacterial capacity against MRSA. Compound 4a showed low toxicity and did not cause drug-resistance in MRSA after multiple generations. Its mechanism of action involves infiltration of the MRSA membrane and insertion into MRSA DNA to prevent replication, resulting in strong inhibition of the bacteria. The highly active derivative 4a also responded well to inflammatory factors and had the potential to be a multifunctional anti-MRSA agent.

Coupling Between Hippocampal Parenchymal Fraction and Cortical Grey Matter Atrophy at Different Stages of Cognitive Decline

An Effective Co-Support Guided Analysis Model for Multi-Contrast MRI Reconstruction

Summary: Multi-contrast magnetic resonance imaging (MRI) is commonly used for clinical diagnosis, but obtaining multi-contrast MR data is time-consuming and may result in motion artifacts. To improve image quality in limited acquisition time, we propose a model that reconstructs images from under-sampled k-space data using fully-sampled data from another contrast of the same anatomy. The model incorporates similarity regularization of the co-supports across multi-contrasts, leading to a mixed integer optimization problem. Experimental results show that our model outperforms other state-of-the-art methods in terms of quantitative metrics and visual performance at various sampling ratios.

Using 2-dimensional hand photographs to predict postoperative biochemical remission in acromegaly patients: a transfer learning approach

Summary: Monitoring postoperative biochemical levels in acromegaly patients, especially those in developing countries, is a challenge. In this study, a mobile and low-cost method using MobileNetv2-based transfer learning algorithm was established to predict biochemical remission. The algorithm showed high accuracy in predicting biochemical remission in both the training and validation cohorts.

Gut Microbiota and Metabolome Changes in Three Pulmonary Hypertension Rat Models

Summary: Dysbiosis of gut microbiota and metabolites is found in pulmonary hypertension patients and rodent models. The exact changes in gut microbiota during the development of pulmonary hypertension remain unclear, as does the function of gut microbiota. This study demonstrated disrupted gut microbiota in rats with various induced forms of pulmonary hypertension, and showed dynamic changes in the gut microbiota during disease progression. Specific plasma biomarkers and a disease-associated network linking Streptococcus with pulmonary hypertension-associated metabolites were identified.

The protective role of EP300 in monocrotaline-induced pulmonary hypertension
