CAIX forms a transport metabolon with monocarboxylate transporters in human breast cancer cells

Tumor cells rely on glycolysis to meet their elevated demand for energy. Thereby they produce significant amounts of lactate and protons, which are exported via monocarboxylate transporters (MCTs), supporting the formation of an acidic microenvironment. The present study demonstrates that carbonic anhydrase IX (CAIX), one of the major acid/base regulators in cancer cells, forms a protein complex with MCT1 and MCT4 in tissue samples from human breast cancer patients, but not healthy breast tissue. Formation of this transport metabolon requires binding of CAIX to the Ig1 domain of the MCT1/4 chaperon CD147 and is required for CAIX-mediated facilitation of MCT1/4 activity. Application of an antibody, directed against the CD147-Ig1 domain, displaces CAIX from the transporter and suppresses CAIX-mediated facilitation of proton-coupled lactate transport. In cancer cells, this metabolon disruption results in a decrease in lactate transport, reduced glycolysis, and ultimately reduced cell proliferation. Taken together, the study shows that carbonic anhydrases form transport metabolons with acid/base transporters in human tumor tissue and that these interactions can be exploited to interfere with tumor metabolism and proliferation.

Summary: Neuronal function requires efficient transport of nutrients into the CNS with protection from unwanted solutes. The Drosophila BBB is formed by specific glial cells that separate the brain from the hemolymph, facilitating the uptake of glucose and trehalose. Loss of BBB transporters MFS3 and Pippin results in pupal lethality, but compensation through upregulation of Tret1-1 rescues transport efficiency and lethality, showing differences between RNA interference and null mutation compensatory mechanisms.

Summary: Intra- and extracellular pH regulation is crucial for cells and tissues, with distorted pH regulation in tumor tissues creating a hostile environment for cancer cells. Cancer cells manipulate a variety of transporters and carbonic anhydrases to alter pH values, promoting tumor progression. Transport metabolons formed by these components play a key role in tumor pH regulation and potential therapeutic interventions for cancer.

Experimental Approaches to Identify Selective Picomolar Inhibitors for Carbonic Anhydrase IX

Summary: This review presents the design, development, and identification of selective nano-to picomolar CA IX inhibitors VD11-4-2, VR16-09, and VD12-09. Various methods and results were discussed, including compound binding, biophysical techniques, protein cloning and purification strategies, and cancer cell-based assays which confirmed nanomolar activities of lead inhibitors. Novel CA IX inhibitors show promise for in vivo exploration and targeting multiple proteins involved in tumor acidosis may improve cancer management.

Summary: This study designed a series of high affinity and high selectivity fluorescein-labeled compounds targeting CAIX for visualization and quantification of CAIX expression in cancer cells. The competitive binding model was used to determine the dissociation constants of common CA inhibitors for CAIX in cancer cells.
