Summary: Intravenously administered iron-carbohydrate preparations are nanomedicines with varying physicochemical characteristics that affect the biodistribution of iron. A systematic comparison of iron exposure and tissue biodistribution among four intravenous iron-carbohydrates in anemic rats was conducted. The study found significant differences in tissue iron biodistribution between the different complexes, suggesting that using serum iron profiles as a surrogate for tissue biodistribution may be erroneous.

Summary: The study explores the application of nanomedicine in the treatment of HIV/AIDS. A combination of gold nanoparticles and Tenofovir was successfully synthesized and showed multifunctional activity against HIV1 and HIV1 protease. The findings highlight the prospects of developing AuNP-TNF as a novel next-generation platform to treat HIV/AIDS.

Summary: Thalassemia is a monogenic autosomal recessive disorder caused by mutations, which affect hemoglobin production. Conventional therapeutic methods such as transfusion and iron chelation therapy have limitations, leading to the demand for advanced therapies. Nanotechnology-based applications have emerged as simple, convenient, and cost-effective methods for theragnostics. This review summarizes the potential of nanotechnology in developing theragnostic approaches for diagnosing thalassemia-causing gene mutations and employing nano-based therapeutic procedures.

Summary: In mouse models, inhibition of TGF beta signaling in combination with checkpoint blockade has been shown to enhance and sustain antitumor immune response. To improve the therapeutic index, tumor-activated prodrugs of a selective small molecule TGF beta R1 inhibitor were developed, selectively metabolized in tumor tissues and reducing systemic exposure of the active drug.

Summary: This paper reports the Fe3+ chelation and therapy of iron overload nonalcoholic fatty liver disease (NAFLD) using fluorescent egg white-based carbon dots (EWCDs) obtained through microwave-assisted pyrolysis method. The high-sensitivity sensor EWCDs show a high correlation between fluorescence emission and Fe3+ concentration in low concentration ranges, with high biocompatibility and specific binding of Fe3+ both in vivo and in vitro. As a novel nano-iron-chelator, EWCDs can successfully attenuate the production of lethal reactive oxygen species, prevent hepatocyte apoptosis, regulate fatty acid metabolism, and alleviate inflammation. Ultimately, they alleviate NAFLD induced by iron overload in zebrafish.

The leading cause of post-LT death is malignancy.

Summary: The etiological investigation of hyperferritinemia involves clinical examination and biological tests. Dysmetabolic iron overload syndrome is the most common iron overload disease, and the effectiveness of bloodletting therapy is uncertain. Type 1 genetic hemochromatosis results in iron accumulation, but phenotypic expression varies. Elastography can be used to identify hepatic fibrosis in cases of elevated ferritin levels. Chelators are used for iron overload associated with anemia, with hepatic iron content needing to be controlled. Deferoxamine is the preferred treatment for severe iron overload or emergency conditions.

Summary: This study utilized small amplitude oscillatory rheometry to investigate the dynamics of blood clot formation upon heparin neutralization under three different oscillatory frequencies, revealing significant differences in blood clot structure formed at a frequency mimicking physiological heart rate. Protamine showed a limited working dose range and intrinsic anticoagulation behavior, while the neutralization profile of UHRA-7 demonstrated a large window of activity. The global assessment of rheological parameters and clot microstructure provided additional details on anticoagulant reversal and blood coagulation dynamics.
