Structural characterization and α-glycosidase inhibitory activity of a novel polysaccharide fraction from Aconitum coreanum

alpha-Glycosidase is an essential target for the management of postprandial serum glucose in diabetic patients. Therefore, the interest has been growing in the screening of alpha-glycosidase inhibitor from natural resource. In the present study, the structure and alpha-glycosidase inhibitory activity of a polysaccharide (named as ACPP-1) from Aconitum coreanum were investigated. Based on the results from high performance gel permeation chromatography, GC-MS and 1D/2D nuclear magnetic resonance spectroscopy, ACPP-1 was a highly-linear polysaccharide with a molecular weight of 34.0 kD and containing over 90 % of glucose. It was composed of (1 -> 4)-alpha-D-Glcp and alpha-Aral. ACPP-1 exhibited a dose-dependent inhibitory effect against alpha-glycosidase activity in vitro and the IC(50 )value was similar to 0.8 mg/mL. In oral starch tolerance test, treatment with ACPP-1 (800 mg/kg) significantly improved the starch tolerance in mice. Taken together, this study provided a potential intervention and management for postprandial hyperglycemia by the polysaccharide fraction from A. coreanum.

Rapid and comprehensive quality assessment of Bupleuri Radix through near-infrared spectroscopy combined with chemometrics

Summary: This study developed a comprehensive strategy based on near infrared spectroscopy and chemometrics for quality assessment of Bupleuri Radix. The strategy successfully identified the geographical origin of Bupleuri Radix and quantified the vital quality indicators through calibration models. Comparison with other models showed that the CARS-SVM model performed better, demonstrating the potential of NIR spectroscopy and appropriate chemometrics for rapid quality evaluation. The developed strategy could supplement existing methods for analysis of Bupleuri Radix and provide reference for quality evaluation and control of other Chinese herbal medicines.

Pyropia yezoensis porphyran alleviates metabolic disorders via modulating gut microbiota in high-sucrose-fed Drosophila melanogaster

Summary: The study found that PYP can improve metabolic abnormalities caused by high sucrose by modulating gut microbiota. PYP reduced the relative abundance of bacteria causing metabolic abnormalities and increased the relative abundance of beneficial bacteria. However, PYP had no effect on triglyceride and circulating sugar contents in larvae fed with high sucrose after gut microbiota removal by antibiotics.

Biodegradation of polybutylene adipate-co-terephthalate by Priestia megaterium, Pseudomonas mendocina, and Pseudomonas pseudoalcaligenes following incubation in the soil

Summary: This study found that a mixture of Pr. megaterium and Ps. mendocina was highly effective at biodegrading PBAT, resulting in approximately 84% weight loss of PBAT film after eight weeks of soil incubation. Mixtures of the other two species also positively affected the synergistic degradation of PBAT film in the soil, but the mixture of three species had a negative effect. The results highlight the importance of lipase secretion in the degradation process of PBAT in the soil.

Near-infrared spectroscopy as a tool to assist Sargassum fusiforme quality grading: Harvest time discrimination and polyphenol prediction

In this study, a high-throughput intact N-glycopeptides quantification strategy was developed, allowing the validation of 20 samples per run with an average analysis time of 3 minutes per sample. The strategy was applied in a cohort of 461 serum samples and identified a panel of 10 IgG N-glycopeptides that have strong clinical utility in evaluating the severity of liver disease.
