Iron-depletion promotes mitophagy to maintain mitochondrial integrity in pathogenic yeast Candida glabrata

Candida glabrata, a haploid budding yeast, is the cause of severe systemic infections in immune-compromised hosts. The amount of free iron supplied to C. glabrata cells during systemic infections is severely limited by iron-chelating proteins such as transferrin. Thus, the iron-deficiency response in C. glabrata cells is thought to play important roles in their survival inside the host's body. In this study, we found that mitophagy was induced under iron-depleted conditions, and that the disruption of a gene homologous to ATG32, which is responsible for mitophagy in Saccharomyces cerevisiae, blocked mitophagy in C. glabrata. The mitophagic activity in C. glabrata cells was not detected on short-period exposure to nitrogen-starved conditions, which is a mitophagy-inducing condition used in S. cerevisiae. The mitophagy-deficient atg32 mutant of C. glabrata also exhibited decreased longevity under iron-deficient conditions. The mitochondrial membrane potential in Cgatg32 cells was significantly lower than that in wild-type cells under iron-depleted conditions. In a mouse model of disseminated infection, the Cgatg32 strain resulted in significantly decreased kidney and spleen fungal burdens compared with the wild-type strain. These results indicate that mitophagy in C. glabrata occurs in an iron-poor host tissue environment, and it may contribute to the longevity of cells, mitochondrial quality control, and pathogenesis.

This study reviewed cases of breakthrough fungemia among adult patients with hematological diseases between 2008 and 2019. The results demonstrate the clinical characteristics and mortality factors associated with breakthrough fungemia in this patient population.

Prevalence of Antifungal Resistance, Genetic Basis of Acquired Azole and Echinocandin Resistance, and Genotyping of Candida krusei Recovered from an International Collection

Summary: This study evaluated the prevalence of antifungal resistance, genetic mechanisms associated with resistance, and genotyping of Candida krusei. The results showed that most isolates were susceptible to most drugs, but a small portion showed resistance. Overexpression of ABC1 gene and FKS1 HS1 mutations were the main mechanisms of resistance. Genotyping confirmed the existence of two main subpopulations.

Summary: There are limited studies on the clinical course of rare/cryptic species-induced invasive aspergillosis (IA) in allo-HSCT recipients. In this retrospective study, it was found that more than half of the proven IA cases were caused by rare/cryptic species. Surgical treatment could be an important option for the management of IA caused by these rare/cryptic species.

Biofilm-formation capability depends on environmental oxygen concentrations in Candida species

Summary: This study investigates the biofilm-forming capabilities of Candida species under different oxygen conditions. The results show that C. albicans favors aerobic conditions to form biofilms, while C. tropicalis demonstrates higher biofilm formation ability and promotes hyphal growth under microaerobic conditions.

Quantitative Monitoring of Mycelial Growth of Aspergillus fumigatus in Liquid Culture by Optical Density

Summary: Filamentous fungi, such as Aspergillus fumigatus, form multicellular hyphae that are difficult to monitor quantitatively in liquid cultures. In this study, we successfully developed a method to quantitatively monitor hyphal growth in A. fumigatus by measuring optical density. This method can be used to establish a standard for hyphal quantification and to screen for novel antifungal agents against Aspergillus species.

Aureobasidium melanigenum catheter-related bloodstream infection: a case report

PCR, along with histopathological diagnosis, is valuable for precise identification, but has limitations in autopsy FFPE samples.

Otitis externa caused by Malassezia slooffiae complicated with mastoiditis: A case report

Summary: Herein, we present a case of otitis externa caused by Malassezia slooffiae complicated with mastoiditis. The patient had fever and severe otorrhea, and was diagnosed with otitis media and mastoiditis based on clinical manifestations and imaging findings. The infection was successfully treated with liposomal amphotericin B and voriconazole.

Promising whole-cell vaccines against cryptococcosis

Summary: Cryptococcosis is a potentially lethal fungal infection caused by Cryptococcus neoformans and C. gattii species complexes. Currently, there is no commercially available prophylactic vaccine and the protective immunity is still under investigation. Whole-cell vaccines, including modified highly immunostimulating strains, have shown effectiveness in prolonging life and suppressing fungal burden in mice, and cross-reactivity to other fungal pathogens. These findings provide valuable insights for vaccine development. In this review, we discuss the advantages and disadvantages of whole-cell vaccines against cryptococcosis.
