Dafachronic acid promotes larval development in Haemonchus contortus by modulating dauer signalling and lipid metabolism

Here, we discovered an endogenous dafachronic acid (DA) in the socioeconomically important parasitic nematode Haemonchus contortus. We demonstrate that DA promotes larval exsheathment and development in this nematode via a relatively conserved nuclear hormone receptor (DAF-12). This stimulatory effect is dose- and time-dependent, and relates to a modulation of dauer-like signalling, and glycerolipid and glycerophospholipid metabolism, likely via a negative feedback loop. Specific chemical inhibition of DAF-9 (cytochrome P450) was shown to significantly reduce the amount of endogenous DA in H. contortus; compromise both larval exsheathment and development in vitro; and modulate lipid metabolism. Taken together, this evidence shows that DA plays a key functional role in the developmental transition from the free-living to the parasitic stage of H. contortus by modulating the dauer-like signalling pathway and lipid metabolism. Understanding the intricacies of the DA-DAF-12 system and associated networks in H. contortus and related parasitic nematodes could pave the way to new, nematode-specific treatments.

Summary: Genomic data provides insights into pest management, resistance evolution, pest invasions, and population dynamics. The first reference genome for the redlegged earth mite, Halotydeus destructor, was assembled to study pesticide adaptation and population demographics using whole-genome pool-seq data. Genetic mutations have been identified that contribute to organophosphate and pyrethroid resistance. Population genomic analyses reveal regional structuring in H. destructor populations, while demographic analyses indicate reduced genetic diversity and smaller effective population sizes in eastern populations. These findings have important implications for genetic markers, understanding resistance mechanisms, and predictive modeling of resistance evolution and spread.

TripletCell: a deep metric learning framework for accurate annotation of cell types at the single-cell level

Summary: Single-cell RNA sequencing (scRNA-seq) has greatly facilitated the characterization of cell lineages and types in complex tissues and organisms. Cell-type annotation is important in scRNA-seq analysis, but manual annotation can be time-consuming and subjective. To address these challenges, we propose a new pipeline called TripletCell for cross-species, cross-protocol, and cross-sample cell-type annotation. Experimental results demonstrate that TripletCell outperforms existing methods and can accurately annotate different cell types regardless of protocols or species.

PI3Kα Translocation Mediates Nuclear PtdIns(3,4,5) P3 Effector Signaling in Colorectal Cancer

Summary: The nuclear translocation of PI3K alpha regulates the subcellular levels of PtdIns(3,4,5)P3 in colorectal cancer cell lines, and 867 potential nuclear PtdIns(3,4,5)P3 effector proteins have been identified. These effector proteins are mainly involved in RNA metabolism and may play a role in modulating pre-mRNA splicing. These findings suggest that nuclear PI3K alpha signaling through PtdIns(3,4,5)P3 effector proteins is important for CRC development.

Summary: Excitotoxicity is a neuronal death process initiated by over-stimulation of glutamate receptors. Proteolytic processing of proteins, particularly by calpains, plays a critical role in this process. Our study identified key synaptic regulatory proteins as substrates of calpains, and their proteolysis contributes to neuronal death by disrupting synaptic organization and function. Blocking calpain-mediated proteolysis of Src protein showed neuroprotective effects. Our findings provide new insights into excitotoxic neuronal death mechanisms and potential targets for neuroprotection.

Structural insights into the multifunctionality of rabies virus P3 protein

Summary: Viruses interact extensively with host proteins to modulate the infected cell's biology, often through multifunctional viral proteins. Summary: Many parasitic worms have a harmful impact on human and animal populations globally. Extracellular vesicles (EVs) have been found to play a role in modulating inflammatory/immune host responses and parasitism. In this study, EVs from the Haemonchus contortus parasite were enriched and a comprehensive analysis of EV proteins and lipids was conducted. The findings provide insights into the role of EV-specific molecules in parasite-host cross-talk and offer potential strategies to disrupt or eliminate parasite infection.

Summary: Biodiversity within the animal kingdom is associated with extensive molecular diversity. The expansion of genomic, transcriptomic, and proteomic data sets for invertebrate groups necessitates reliable in silico tools for molecular identification. This study established a sequence- and structure-based workflow system to identify and annotate proteases and protease inhibitors of a highly pathogenic parasitic roundworm. The results provide a solid foundation for further exploration of these molecules and their potential as drug targets. Rating: 8/10.

This discovery provides new insights for designing durable antifungal surfaces to prevent biofilm formation by pathogenic, multi-drug resistant yeasts.
