Nitrite induces endoplasmic reticulum stress and associates apoptosis of liver cells in grass carp (Ctenopharyngodon idella)

Nitrite pollution is one of the most frequently reported problems in aquaculture. However, little is known for its toxic mechanisms. The present study was conducted to investigate the effects of nitrite on antioxidant capacity, ER stress (Endoplasmic Reticulum stress) and apoptosis in grass carp (Ctenopharyngodon idella). Fish were exposed to nitrite (0, 8, 25, 50 mg L-1) for 96 h, the genes expression, histological section and antioxidant enzyme activity were assayed. The results show that the expression of ER stress marker gene grp78 was significantly upregulated in liver, the expression of PERK pathway genes perk, eif-2 alpha, atf4 and chop have changed, the superoxide dismutase (SOD) activity decreased while the catalase (CAT) activity increased significantly in serum, fish liver shows severe damage in nitrite exposure. Grass carp liver cells (L8824) were incubated in nitrite solution (0, 2.5, 10, 100 mg L-1) for 24 h. Significant decrease in cell viability and apoptosis were observed in the treatment, the expression of ER stress marker gene grp78 was up-regulated, moreover the PERK pathway genes perk, eif-2 alpha, atf4 and chop were up-regulated in 2.5 mg L-1 and 10 mg L-1. The expression of the proapoptotic gene bax was significantly up-regulated, and the expression of the inhibitory gene bcl-2 was significantly down-regulated, therefore the ratio of bax/bcl-2 was up-regulated. These results indicate that in the nitrite-induced acute liver injury of grass carp, ER stress response may activate antioxidant system as a protection.

Summary: A new in-pond tank culture system (IPTCS) based on the concept of pond partitioned aquaculture system was constructed in this study. The experiment focused on high and low stocking densities of grass carp and blunt snout bream. Results showed that high stocking density had a negative impact on growth performance and muscle quality, as well as water quality in the culture tank. Economic analysis revealed that high-density rearing of blunt snout bream led to higher income.

Protective Effect of Selenium on the Oxidative Damage of Kidney Cells Induced by Sodium Nitrite in Grass Carp (Ctenopharyngodon idellus)

Summary: The present study demonstrated that selenium supplementation can enhance the resistance of cells to sodium nitrite exposure by alleviating the antioxidant imbalance, high apoptosis rate, and intracellular calcium ion disturbance caused by nitrite exposure. Pre-incubation of CIK cells with selenium maintained antioxidant indicators at normal levels and decreased intracellular calcium ion concentration and apoptotic rate compared to nitrite exposure. The Nrf2-Keap1 pathway may play an important role in mediating the protective effects of selenium against nitrite-induced oxidative damage.

The growth performance, antioxidative status and muscle quality of grass carp (Ctenopharyngodon idellus) cultured in the recirculating pond aquaculture system (RPAS)

Summary: This study explored the performance of the recirculating pond aquaculture system (RPAS) on grass carp. The results showed that RPAS had lower concentrations of nitrogen compounds in the water and led to higher growth rates and lower feed conversion rates in the fish. The expression of growth-related genes was also increased in the fish cultured in RPAS ponds. Additionally, RPAS helped reduce oxidative stress and enhance muscle umami flavor in the fish.

Formation of microorganism-derived dissolved organic nitrogen in intermittent aeration constructed wetland and its stimulating effect on phytoplankton production: Implications for nitrogen mitigation

Summary: To control eutrophication in aquatic ecosystems, upgrading conventional constructed wetlands to aeration wetlands is common. However, efforts to reduce dissolved inorganic nitrogen (DIN) discharge have overshadowed the importance of dissolved organic nitrogen (DON). This study found that intermittent aeration in aeration wetlands introduces large amounts of DON and bioavailable DON in effluents, while decreasing total nitrogen (TN). The increase in DON discharge can influence phytoplankton composition and stimulate harmful species, indicating the need to regulate DON discharge alongside DIN.

Assessment of Aquatic Ecological Health Based on the Characteristics of the Fish Community Structures of the Hun River Basin, Northeastern China

Summary: Long-term ecological restoration can partially restore aquatic ecosystems and alleviate the crisis of freshwater fish biodiversity. The distribution of fish communities in the Hun River Basin showed spatial differences along the longitudinal gradient and was influenced by agricultural land, urban land, and grassland. The health status of the Hun River varied, with the upper and middle reaches being in better health and the lower reaches being poorer, mainly due to different levels of human activities.

Comparative Analysis of Metabolites between Different Altitude Schizothorax nukiangensis (Cyprinidae, Schizothoracine) on the Qinghai-Tibet Plateau in Nujiang River

Summary: Metabolomics studies were used to investigate the influence of the high-altitude aquatic environment on indigenous fish metabolites. Different metabolite concentrations were found in the livers of fish at high and low altitudes, indicating different metabolic processes such as oxidative stress, energy metabolism, carbohydrate metabolism, and lipid metabolism. The mTOR signaling pathway, apoptosis, and lysosome were enriched between groups to ensure energy supply and limit tissue damage in fish at high altitudes. Nicotine and methoprene, two organic pollutants, showed different effects in fish at different altitudes. Overall, this study provides important insights into fish adaptation to high-altitude environments and organic contaminants pollution.

Longitudinal Patterns in Fish Assemblages after Long-Term Ecological Rehabilitation in the Taizi River, Northeastern China

The findings provide valuable perspectives for species conservation.

Acute Thermal Stress Reduces Skeletal Muscle Growth and Quality in Gibel Carp (Carassius gibelio)

Summary: This study examined the effects of acute thermal stress on skeletal muscle growth and quality in gibel carp. Results showed that the muscle nutrient content was significantly higher at 20°C, with the muscle being more tender and elastic. The gene expression levels of the MRFs family were upregulated after 1 hour and gradually decreased. The study suggests that gibel carp can adapt to acute thermal stress to some extent, but it eventually leads to a decrease in muscle growth rate and quality.

Diet-egg relationships and equilibration of fatty acids in eggs of southern flounder, Paralichthys lethostigma

Evaluation of machine learning method in genomic selection for growth traits of Pacific white shrimp
