Summary: A series of high-resolution cryo-EM structures of substrate-bound human p97 AAA ATPase revealed critical conformational changes and 'power-stroke'-like motions in the ATPase rings, shedding light on the central role of p97 in cellular protein homeostasis. The cryo-EM structure of p97 complexed with the inhibitor NMS-873 at a resolution of 2.4 angstrom demonstrated the mechanism of allosteric inhibition by blocking substrate translocation.

Summary: Ulcerative colitis (UC) is a chronic inflammatory disease of the colon characterized by immune dysregulation. Pre-treatment with TNF-alpha and IFN-gamma enhances the therapeutic potential of human amniotic epithelial cells (hAECs) in treating UC. Pre-hAECs were found to be more effective in alleviating colitis in mice models compared to hAECs alone. The treatment with pre-hAECs resulted in reduced weight loss, shortened colon length, decreased disease activity index, and maintained the recovery of colon epithelial cells.

Summary: The expression of IL-10 in inflamed dental pulp and its effects on the plasminogen activation system molecules have been investigated. Melatonin, a physiological endocrine generated by the pineal gland, was found to have anti-fibrinolytic effects on IL-10-induced changes in uPA, uPAR, and PAI-1 in dental pulp cells. This research suggests that melatonin and signal transduction inhibitors may be used for the prevention and treatment of pulpal inflammatory diseases.
