Summary: This paper discusses the pathologic research and drug therapy of diabetic kidney disease (DKD), focusing on the death processes and mechanisms of glomerular podocytes in DKD. The complexity and potential crosstalk between various modes of cell death are also examined, providing insights into podocyte death and laying the foundation for future targeted therapy strategies for DKD treatment.

Summary: Klotho activates the Nrf2 signaling pathway to inhibit oxidative stress and apoptosis induced by high glucose, thus protecting podocytes. This protective effect may be targeted by activating Nrf2 for the treatment of diabetic nephropathy (DN).

Summary: The combination of metformin and venetoclax has been found to effectively inhibit the growth of AML cells both in vitro and in vivo. This combination treatment induces cell apoptosis and increases the expression levels of the endoplasmic reticulum stress-related marker CHOP, which plays a crucial role in the anti-leukemic activity.

Summary: Arsenic is the primary toxicant among hazardous environmental chemicals, implicated in neurotoxicity such as cognitive dysfunction, neurodevelopmental changes, and neurodegenerative diseases. It is believed that sustained pro-inflammatory response may trigger arsenic-induced neurotoxicity, potentially involving microglia activation and release of pro-inflammatory cytokines. Further research is needed to clarify the role of these cytokines in arsenic-induced neurotoxicity.

Summary: In diabetic kidney disease (DKD), impaired ER-phagy of renal tubular cells worsens tubular injury. It has been found that the deficiency of PACS-2 aggravates tubular injury in DKD by inhibiting ER-phagy via the TFEB/FAM134B pathway.

Summary: Intermedin (IMD) has renoprotective effects on diabetic nephropathy by attenuating podocyte apoptosis and restoring slit diaphragm protein deficiency. The mechanism involves blocking endoplasmic reticulum stress responses in renal podocytes, reducing apoptosis and promoting slit diaphragm protein synthesis.

Summary: The endoplasmic reticulum (ER) is an active organelle involved in various physiological processes. Recent studies have shown that dysfunction of the ER plays a role in the progression of kidney disease, particularly diabetic nephropathy (DN). This review discusses the function of the ER, the regulation of homeostasis through the unfolded protein response (UPR) and ER-phagy, as well as the impact of abnormal ER homeostasis in renal cells in DN. Additionally, potential therapeutic targets for DN related to maintaining ER homeostasis are also explored.

Summary: The study showed that ChREBP deficiency can improve the expression of TXNIP in patients with diabetic nephropathy, leading to improved renal function, apoptosis, and ER stress. Additionally, ChREBP deficiency can prevent the increased expression of TXNIP, Nox4, 8-OHdG, and HO-1 in diabetic kidneys, suggesting ChREBP as a potential therapeutic target for diabetic nephropathy.

Summary: This study investigated the molecular mechanisms of beta-arrestin2 in primary Sjogren's syndrome, finding that beta-arrestin2 downregulates GRP78-ATF6-CHOP apoptosis signaling to improve symptoms and tissue integrity in an experimental Sjogren's syndrome mouse model. The research suggests that excessive activation of GRP78-ATF6-CHOP apoptosis signaling is involved in the pathogenesis of primary Sjogren's syndrome, and beta-arrestin2 encourages inflammation-induced epithelial apoptosis through GRP78-ATF6-CHOP apoptosis signaling.
