Lymphoid Aggregates Remodel Lymphatic Collecting Vessels that Serve Mesenteric Lymph Nodes in Crohn Disease

Early pathological descriptions of Crohn disease (CD) argued for a potential defect in Lymph transport; however, this concept has not been thoroughly investigated. In mice, poor healing in response to infection-induced tissue damage can cause hyperpermeable lymphatic collecting vessels in mesenteric adipose tissue that impair antigen and immune cell access to mesenteric lymph nodes (LNs), which normally sustain appropriate immunity. To investigate whether analogous changes might occur in human intestinal disease, we established a three-dimensional imaging approach to characterize the Lymphatic vasculature in mesenteric tissue from controls or patients with CD. In CD specimens, B-cell rich aggregates resembling tertiary lymphoid organs (TLOs) impinged on lymphatic collecting vessels that enter and exit LNs. In areas of creeping fat, which characterizes inflammation-affected areas of the bowel in CD, we observed B cells and apparent innate lymphoid cells that had invaded the lymphatic vessel wall, suggesting these cells may be mediators of Lymphatic remodeling. Although TLOs have been described in many chronic inflammatory states, their anatomical relationship to preeStablished LNs has never been revealed. Our data indicate that, at least in the CD-affected mesentery, TLOs are positioned along collecting lymphatic vessels in a manner expected to affect delivery of lymph to LNs.

Summary: The development of nanotechnologies has led to an increase in engineered nanoparticles (EN) in the environment, including the atmosphere. This study assessed the impact of four prevalent atmospheric EN (silver, titanium dioxide, titanium, and silicon dioxide) on intestinal inflammation and microbiota. The results suggest that these EN can disrupt intestinal homeostasis by modulating cytokine expression and altering the inflammatory response and microbiota composition.

Summary: Recent studies have shown that the skin acts as a major reservoir for sodium ions by regulating the content of glycosaminoglycans and osmotic gradients. This study investigated the presence of electrolyte gradients in the skin and the storage of sodium ions from a fluid balance perspective. The results revealed a gradient of sodium ions from the epidermis to the dermis, with a higher concentration in the epidermis. This hydration-dependent concentration of sodium ions in the interstitial fluid contributes to the skin barrier and limits water loss. Additionally, intracellular storage of sodium ions in muscle and myocardium may have electromechanical consequences in salt stress.

Summary: The aim of this study was to investigate the effects of intestinal exposure to polyethylene (PE) microplastics on gut homeostasis. Mice were orally exposed to PE microbeads of two different sizes, and it was found that ingestion of PE microbeads led to intestinal disturbances and significant alterations of crucial intestinal markers. This highlights the need for further research on the health impact of microplastic exposure in humans.

Summary: The transcription factor Basonuclin 2 (BNC2) is found to play a key role in driving myofibroblast activation in liver fibrosis. Its expression is induced in fibrotic livers and it integrates pro-fibrotic stimuli to induce matrisome gene expression. BNC2 deficiency leads to reduced collagen deposition in the liver. Furthermore, the study identifies a potential drug target, CC-885, as a BNC2 inhibitor.

In vitro and in vivo evaluation of a pH-, microbiota- and time-based oral delivery platform for colonic release

Summary: A novel drug delivery system based on multiple drug release triggers was developed and evaluated for oral colon delivery, with a specific focus on inflammatory bowel disease (IBD) therapy. The system demonstrated pulsatile drug release in different release media and faster drug release in simulated colonic fluid with fecal bacteria from IBD patients. Promising results were observed in a preliminary rat study, indicating potential therapeutic effects on colitis progression and microbiota alteration.

Starch nanoparticles improve curcumin-induced production of anti-inflammatory cytokines in intestinal epithelial cells

Summary: This study investigated the use of crosslinked starch nanocarrier (NPL) on the intracellular delivery and anti-inflammatory efficiency of curcumin in inflammatory bowel disease (IBD). The results showed that NPL enhanced the intracellular delivery of curcumin in inflammatory cells and decreased the secretion of pro-inflammatory cytokines while increasing the secretion of anti-inflammatory cytokines. Additionally, NPL/Cur exhibited better paracellular transport across the epithelium and showed potential as a therapeutic alternative for IBD management.

An extended transcription factor regulatory network controls hepatocyte identity

Summary: Cell identity is controlled by a core transcriptional regulatory circuitry (CoRC) consisting of cell-specific transcription factors (TFs). However, our study reveals that the transcriptional regulatory network regulating hepatocyte identity is more complex than initially thought. We identified a set of TFs, which we call hepatocyte identity (Hep-ID)(CONNECT) TFs, that not only control hepatocyte identity effector genes, but also interact with TFs of the CoRC. Furthermore, we found that Hep-IDCONNECT TFs play a role in fine-tuning CoRC TF expression and can reset CoRC TF expression in dedifferentiated hepatocytes.
