Gas chromatography coupled to a quadrupole time-of-flight mass analyzer (QTOF) with an atmospheric pressure chemical ionization (APCI) source has been tested to study the ionization and mass spectrometric behavior of long-chain and very long-chain polyunsaturated fatty acids (LC-PUFAs, C18-24; VLC-PUFAs, >C-24). The protonated molecule ([M+H](+)), measured at accurate mass, became the base peak of the spectrum for all the studied compounds and was promoted by the addition of water into the source. This fact overcame the existing difficulties for the identification of VLC-PUFAs when using an electron ionization source (EI). The extensive fragmentation of PUFAs in this source is the main drawback due to the fact that since reference standards are not commercially available, final identification relies on retention time estimation. The application of GC-APCI-QTOF to the screening of lipid extracts from the eyes of different fish species added confidence to the identification of several VLC-PUFAs. Further investigation of ion ratios allowed to predict the position of key double bonds enabling the classification of VLC-PUFAs as omega 3 or omega 6 compounds. VLC-PUFAs spectra found in biological samples were compared to those obtained from corresponding peaks found in heterologous expression experiments of fish's Elovl4. (C) 2018 Elsevier B.V. Identification of new, very long-chain polyunsaturated fatty acids in fish by gas chromatography coupled to quadrupole/time-of-flight mass spectrometry with atmospheric pressure chemical ionization

Roque Serrano, Juan Carlos Navarro, Tania Portoles, Carlos Sales, Joaquin Beltran, Oscar Monroig, Felix Hernandez

Summary: The study developed a new methodology for identifying very long-chain polyunsaturated fatty acids (VLC-PUFAs), successfully discovering VLC-PUFAs with chain lengths up to 44 carbons in gilthead sea bream. The research also evaluated the added value of ion mobility-mass spectrometry and the contribution of collisional cross section parameters in characterizing novel VLC-PUFAs.

Tandem mass spectrometry of organic nitro and halogen compounds: Competition between losses of molecules and of radicals

W. M. A. Niessen

Summary: The discussion focuses on the fragmentation of protonated and/or deprotonated small-molecule compounds in tandem mass spectrometry after electrospray ionization, with a special emphasis on nitro and halogen-related fragmentation. The research indicates that nitro and halogen moieties may lose a radical, and the chemical surroundings can influence the fragmentation process, further affecting the different molecular fragmentation pathways.

Effect of short-term exposure to fluorescent red polymer microspheres on Artemia franciscana nauplii and juveniles

Diogo Peixoto, Amparo Torreblanca, Susana Pereira, Maria Natividade Vieira, Inmaculada Varo

Summary: Microplastics are found everywhere in the world's oceans and their potential toxic effects on aquatic ecosystems remain unknown. This study evaluated biochemical responses caused by fluorescent red polymer microspheres (FRM) on nauplii and juveniles of Artemia franciscana, revealing that juveniles showed decreased survival after exposure to higher concentrations of FRM, potentially indicating neurotoxic and oxidative damage effects induced by the microplastics. The research highlights the need to understand the mechanisms of microplastics toxicity and their impacts on environmental safety.

Summary: Long-chain (>= C-20) polyunsaturated fatty acids (LC-PUFAs) are physiologically important for most animals, including humans. Recent research indicates the ability of certain invertebrates, mainly marine organisms, to synthesize and convert LC-PUFAs. The genetic pathways and mechanisms of LC-PUFA biosynthesis remain unknown in many invertebrates, but existing genomic resources can contribute to our understanding. This study identified key enzymes responsible for LC-PUFA biosynthesis in the copepod Platychelipus littoralis and demonstrated their functionality using heterologous expression in yeast.

New insights into the search of meat quality biomarkers assisted by Orbitrap Tribrid untargeted metabolite analysis and chemometrics

Borja Garlito, Miguel A. Sentandreu, Vicent Yusa, Mamen Olivan, Olga Pardo, Enrique Sentandreu

Summary: Metabolite profiles of normal and defective dry, firm and dark (DFD) meat extracts were analyzed using Orbitrap Tribrid ID-X untargeted analysis coupled to chemometrics. Threonylphenylalanine (overexpressed in normal meats) and tetradecadienoyl- and hydroxydodecanoyl-carnitines (both overexpressed in DFD meats) appropriately grouped meat groups assayed. Results revealed the limitations of pHu as an isolated parameter for accurate meat quality differentiation and suggested interferences induced by pre-slaughter stress (PSS) on lipid catabolism.

Miniaturized ascorbic acid assay platform based on point discharge atomic emission spectrometry coupling with gold filament enrichment

Yanping Wang, Yuemei Chen, Kejun Li, Jinrong Zhou, Xin Yuan, Mei Zhang, Ke Huang

Summary: In this work, a portable analytical system based on point discharge chemical vapor generation atomic emission spectrometry (PD-CVG-AES) coupling with gold filament enrichment was designed. The highly sensitive analysis of Hg2+ indirectly realized the detection of ascorbic acid (AA). The measurement is based on the fact that Ag+ can decrease the concentration of Hg2+ by forming Ag-Hg amalgam in the presence of the reductant SnCl2, while AA can pre-reduce Ag+ to Ag0, leading to the generation of silver nanoparticles (Ag NPs). The developed novel analytical strategy broadens the application of microplasma-based AES and offers a higher level of sensitivity compared to current AA detection techniques.
