Summary: Current mpox outbreaks in non-endemic regions are a global public health concern. Two multi-antigen mRNA vaccine candidates, Rmix4 and Rmix6, were developed using a simplified manufacturing strategy and showed promising results in inducing potent immune responses against the virus. Both candidates protected mice from lethal virus challenge and revealed vulnerability to viral immune evasion.

Beta-propiolactone-inactivated vaccine showed higher protection efficacy against virus challenge compared to formaldehyde, generating specific IgM and neutralizing antibodies against TiLV. The vaccine also significantly lowered viral loads, increased survival rates, and stimulated a protective immune response, indicating potential for inhibiting viral proliferation.

Summary: Infectious pancreatic necrosis (IPN) is a highly contagious disease causing high mortality in juvenile trouts, and early diagnosis and prevention are crucial. In Turkey, where there are no commercial licensed vaccines against IPNV, attempts have been made to produce recombinant VP2 subunit vaccine (IPNV-VP2) and inactivated whole particle virus vaccine (IPNV-WPV) from local isolates. The results showed that IPNV-WPV vaccine stimulates immune response faster and has a higher relative percent of Survive (RPS) compared to IPNV-VP2 vaccine in rainbow trout.

Summary: This study confirms the feasibility of attenuating SARS-CoV-2 through the generation of recombinant viruses lacking two accessory ORF proteins. These double ORF-deficient viruses have reduced replication kinetics and fitness compared to the wild-type virus. Importantly, they show attenuation in both transgenic mice and golden Syrian hamsters, providing protection against SARS-CoV-2 infection and transmission. Vaccination with these attenuated viruses induces strong immune responses and can prevent COVID-19.
