The long-term use of anti-inflammatory drugs is the most common cause of gastric ulcer disease, one of the major gastrointestinal disorders affecting people worldwide. Persea americana Mill. (avocado) seed is a by-product generally discarded as waste, but can be used to treat gastric disorder due to its anti-inflammatory, antioxidant and antimicrobial activities. The aim of the present study was to evaluate the potential protective effects of the ethyl acetate fraction of avocado seeds (SEAP) extracts against indomethacin-induced gastric ulcer in mice. It was found that SEAP were effective in mitigating oxidative stress through a decrease on the oxidized products levels (reduction of 90% in lipid peroxidation in plasma) and increasing superoxide dismutase enzyme (SOD) activity (4.25-fold increase compared to the indomethacin group), also preventing the rise in ulcer and lesions areas (92% of protection) and histological changes induced by indomethacin. Chemical analysis using mass spectrometry by ( - )-ESI-FT-ICR MS revealed the presence of ( - )-epicatechin and ( + )-catechin, confirmed by HPLC-DAD, and other important phenolic compounds in avocado seeds, such as caffeoylquinic acid, flavonoids, phenylpropanoids and tannins, substances that promote inhibition of pathways involved in gastric ulcer formation. Thus, avocado seeds extract may be a suitable natural source for the prevention and treatment of gastric ulcer.

The Potential of Vouacapanes from Pterodon emarginatus Vogel Against COVID-19 Cytokine Storm

Leandra de Almeida Ribeiro Oliveira, Arthur Christian Garcia da Silva, Douglas Vieira Thomaz, Fabiana Brandao, Edemilson Cardoso da Conceicao, Marize Campos Valadares, Maria Tereza Freitas Bara, Damaris Silveira

Summary: The aim of this study was to evaluate the anti-inflammatory activity of P. emarginatus oleoresin and isolated diterpenes in in vitro models. The results showed that oleoresin, V2, and V3 inhibited phospholipase A2 activity, and both V1 and V2 inhibited the expression of COX-2. Meanwhile, oleoresin could also inhibit the production of interleukin 6. In conclusion, the results support the ethnomedicinal use of P. emarginatus oleoresin as an anti-inflammatory herbal medicine, and suggest its potential therapeutic use for COVID-19.

Isaac de Araujo Matos, Ana Carolina Goes Pinto, Matheus Vitor Ferreira Ferraz, Wenny Camilla Santos Adan, Ricardo Pereira Rodrigues, Juliane Xavier dos Santos, Rodrigo Rezende Kitagawa, Roberto Dias Lins, Tiago Branquinho Oliveira, Nivan Bezerra da Costa

Summary: In this study, molecular docking simulations, quantitative structure-activity relationships studies, and ADMETox predictions were used to analyze the molecular recognition and inhibition of Staphylococcus aureus dihydrofolate reductase by a series of 7-aryl-2,4-diaminoquinazoline derivatives. A quantitative structure-activity relationship model was developed, demonstrating the importance of pKa and molar refractivity for inhibitory activity. Virtual screening identified PC-124127620, PC-124127795, and PC-124127805 as promising candidates for SaDHFR inhibitors, with high potency and favorable pharmacokinetics and toxicological profiles. These findings suggest these compounds could be potential antimicrobial agents for treating S. aureus infections.

A New Lignan from Annona squamosa L. (Annonaceae) Demonstrates Vasorelaxant Effects In Vitro

Camilo Di Giulio, Juan Manuel Gonzalez Guzman, Joao Victor Dutra Gomes, Young Hae Choi, Perola O. Magalhaes, Yris M. Fonseca-Bazzo, Damaris Silveira, Omar Estrada

Summary: A new furofuran lignan, esquamosan, was isolated from the leaves of Annona squamosa L. and its structure was determined by spectroscopic methods. Esquamosan showed concentration-dependent inhibition of contraction in rat aortic rings and inhibited vasocontraction of depolarized aorta with high-concentration potassium. The vasorelaxant effect of esquamosan was mainly attributed to the inhibition of calcium influx and increased release of NO. Additionally, esquamosan exhibited antioxidant capacity similar to ascorbic acid.

Chemical profile and cytotoxic evaluation of aerial parts of Euphorbia tirucalli L. on gastric adenocarcinoma (AGS cells)

Larissa Silva de Souza, Cristina Luz Tosta, Jessica Rodrigues Pereira de Oliveira Borlot, Marcia C. B. N. Varricchio, Rodrigo Rezende Kitagawa, Paulo Roberto Filgueiras, Ricardo Machado Kuster

Summary: The ethanol extract from Euphorbia tirucalli L. and its latex suspended in water are used in Brazil for the treatment of various diseases, including cancer. This study aimed to determine the cytotoxic effects of the ethanol extract on gastric adenocarcinoma cells (AGS) and analyze its chemical composition. Hexane and aqueous fractions of the extract showed higher cytotoxicity on AGS cells. Analysis revealed the presence of triterpene compounds in the hexane fraction and ellagic acid as the main phenolic compound in the aqueous extract, suggesting that these components contribute to the greater cytotoxic activity of the ethanol extract compared to latex.
