Chaperone-mediated autophagy (CMA) selectively delivers cytosolic proteins with an exposed CMA-targeting motif to lysosomes for degradation and plays an important role in protein quality control and cellular homeostasis. A growing body of evidence supports the hypothesis that CMA dysfunction may be involved in the pathogenic process of neurodegenerative diseases. Both down-regulation and compensatory up-regulation in CMA activities have been observed in association with neurodegenerative conditions. Recent studies have revealed several new mechanisms by which CMA function may be involved in the regulation of factors critical for neuronal viability and homeostasis. Here, we summarize these recent advances in the understanding of the relationship between CMA dysfunction and neurodegeneration and discuss the therapeutic potential of targeting CMA in the treatment of neurodegenerative diseases.

Opinions and clinical practice of functional movement disorders: a nationwide survey of clinicians in China

Xin-Yi Xie, Guo-zhen Lin, Qiang Huang, Chun-Bo Li, Mark Hallett, Valerie Voon, Ru-jing Ren, Sheng-di Chen, Gang Wang

Summary: The survey found that Chinese clinicians consider atypical movement disorders, multiple somatizations, and emotional disturbance to be essential for the diagnosis of functional movement disorders (FMD). However, lack of guidelines, physician knowledge, and training often limit their ability to manage patients with FMD. Efforts are needed to promote related research and establish practice guidelines in China in the future.

Adult-Onset Neuronal Ceroid Lipofuscinosis With a Novel DNAJC5 Mutation Exhibits Aberrant Protein Palmitoylation

Qiang Huang, Yong-Fang Zhang, Lin-Jie Li, Eric B. Dammer, Yong-Bo Hu, Xin-Yi Xie, Ran Tang, Jian-Ping Li, Jin-Tao Wang, Xiang-Qian Che, Gang Wang, Ru-Jing Ren

Summary: Neuronal ceroid lipofuscinosis (NCL) is a group of inherited neurodegenerative diseases characterized by the presence of lipofuscin deposits in the lysosomal lumen. In this study, a novel C128Y mutation in the DNAJC5 gene was identified in a young Chinese female with adult-onset NCL, leading to abnormal palmitoylation and lipofuscin deposits.

The impacts of health insurance and resource on the burden of Alzheimer's disease and related dementias in the world population

Jin-Tao Wang, Gang Xu, Ru-Jing Ren, Ying Wang, Ran Tang, Qiang Huang, Jian-Ping Li, Murad Al-Nusaif, Wei-Dong Le, Gang Wang

Summary: This study investigates the impact of health insurance and resources on the burden of Alzheimer's disease and related dementias (ADRD). The findings suggest that ADRD has an increasing burden on patients globally, both physically and economically. Moreover, health resources are negatively associated with certain burden factors, such as disability-adjusted life years (DALY) and death, but positively associated with years of life lived with disability (YLD).

Speech silence character as a diagnostic biomarker of early cognitive decline and its functional mechanism: a multicenter cross-sectional cohort study

Summary: Language deficits often occur during the early stages of Alzheimer's disease (AD), and the percentage of silence duration (PSD) has been found to be a reliable diagnostic biomarker for early-stage AD and amnestic mild cognitive impairment (aMCI). The brain's response to fluency tasks is significantly inhibited in the aMCI/AD group, which partially explains the elevated PSD.

Genetic profiles of familial late-onset Alzheimer's disease in China: The Shanghai FLOAD study

Xin-Yi Xie, Qian-Hua Zhao, Qiang Huang, Eric Dammer, Sheng-di Chen, Ru-Jing Ren, Gang Wang, Alzheimer's Dis Neuroimaging Initiat

Summary: Compared with early-onset familial Alzheimer's disease, the heritability of most familial late-onset Alzheimer's disease remains unclear. In this study, targeted sequencing of selected candidate genes was conducted for Chinese Han individuals with familial late-onset Alzheimer's disease, revealing the most frequent variants in CR1, TREM2, and ACE. Additionally, novel pathogenic mutations in ACE were found to be associated with familial late-onset Alzheimer's disease.
