Abnormal tumor vasculature and endothelial cell anergy limit tumor/T-cell interactions. We have found that NGR-TNF, a tumor vasculature-homing derivative of TNF, selectively activates endothelial cells in neoplastic tissues and induces the release of chemokines that favor tumor infiltration by T cells, thereby enhancing the efficacy of active and adoptive immunotherapy.

Summary: The blood-brain tumor barrier (BBTB) is a major obstacle for delivering anticancer drugs to central nervous system tumors. Recent advances in various conjugates and drug delivery technologies have shown promising results in breaching the BBTB and increasing drug delivery to brain tumors, without significant adverse events.

Summary: The gut microbiota enriched for species capable of converting androgen precursors into active androgens was found to contribute to the onset of castration resistance in CRPC. Ablation of the gut microbiota by antibiotic therapy delayed the emergence of castration resistance in mice. Fecal microbiota transplantation from CRPC patients rendered mice resistant to castration.

Targeting the Blood-Brain Tumor Barrier with Tumor Necrosis Factor-α

Summary: The blood-brain tumor barrier is a major obstacle for the delivery of anticancer drugs to brain tumors. This review focuses on the development of a drug called NGR-TNF that can target and alter the blood-brain tumor barrier. Preclinical studies have shown that NGR-TNF is a promising strategy for delivering TNF to tumor vessels and breaking the biological barriers. Clinical studies have also demonstrated the efficacy of NGR-TNF in improving patient survival. In addition, the review discusses the challenges and potential solutions for the instability and molecular heterogeneity of NGR-TNF, as well as other peptide/antibody-TNF fusion products and nanoparticles for targeting the blood-brain tumor barrier.

Summary: Tumor cells promote the recruitment of immunosuppressive neutrophils, a subset of myeloid cells driving immune suppression, tumor proliferation, and treatment resistance. We have identified a subset of neutrophils that have upregulated expression of cellular senescence markers and persist in the tumor microenvironment. These senescent-like neutrophils express TREM2 and are more immunosuppressive and tumor-promoting than canonical immunosuppressive neutrophils.

Summary: Tumour-infiltrating myeloid-derived suppressor cells (MDSCs) are a heterogeneous population of myeloid cells that display immunosuppressive activity in tumor patients, especially in hormone-driven tumors.
