Summary: This study suggests that targeting sphingosine kinase 1 (SK1), a key enzyme upregulated in various types of cancer, can effectively decrease tumor growth in lymphoma without functional p53. This positions SK1 as a potential therapeutic target for tumors that lack functional p53.

Summary: DNA is crucial for organism development and function. MiRNA, a type of non-coding RNA, may play an important role in the DNA damage response by influencing processes like cell cycle, DNA repair, and apoptosis, thereby impacting genomic stability and tumorigenesis.

Summary: Heart failure has become an epidemic in an aging population, and DNA damage is believed to play a role in its development. This study found that impaired DNA repair in cardiomyocytes leads to early onset of heart failure, with increased oxidative stress, fibrosis, and apoptosis. The findings suggest that DNA damage could be a potential therapeutic target for heart failure.

Summary: FL enhances the radioresistance of esophageal squamous cell carcinoma cells through promoting clone formation, increasing EdU incorporation, enhancing DNA damage repair, and inhibiting apoptosis. The significantly increased expression of Flt-3 receptor in ESCC cells after radiation may play a key role in their radioresistance.

Summary: Base and prime editors have shown potential for precise genetic engineering, but their cellular responses and genotoxicity are not well understood. This study compared the efficiency, toxicity, and genotoxicity between base and prime editors and Cas9 in human hematopoietic cells. The findings suggest that these editors can induce harmful effects and genotoxicity, raising concerns for their clinical applications.

Summary: Glioblastoma multiforme (GBM) is a highly deadly brain tumor. UBA1 inhibition by TAK-243 can increase the sensitivity of GBM to radiotherapy by impairing DNA double-strand break repair. This study provides a new therapeutic strategy for improving the radiation sensitivity of GBM and lays a foundation for further clinical trials.

Summary: This study reveals the key regulatory role of Mad2 phosphorylation in checkpoint defects and DNA damage repair in ATM-deficient cells. ATM negatively regulates the phosphorylation of Mad2, causing decreased DNA damage repair capacity and resistance to cancer cell radiotherapy.

Summary: Our study investigated whether circadian rhythm affects the DNA damage responses induced by cisplatin. The results showed that the DNA damage responses are regulated by circadian control only in cells with a circadian rhythm, and not in cells without a circadian rhythm. These findings have potential implications for improving or designing chronotherapy approaches for cancer patients.

Summary: In this study, the researchers found that deletion of SphK1 significantly decreased liver damage and mortality rate caused by acute liver failure, likely through regulating the secretion of pro-inflammatory cytokine TNF alpha. The findings suggest that SphK1 may play a crucial role in the development of acute liver failure and could be a potential therapeutic target for fulminant hepatitis.

Summary: Diabetic retinopathy is a common complication of long-term diabetes that can lead to vision loss. The early understanding and prevention of this condition are limited, with no effective methods until later stages. The study used a Nile rat model to examine the transcriptomic changes in retinal vascular development, aiming to better understand the early pathogenesis of diabetic retinopathy.

Summary: This study aims to understand the early pathogenesis of diabetic retinopathy by analyzing whole retinal vascular transcriptomic changes in a Nile rat model. The study identified the complex associations between acellular capillary density and factors like blood glucose, diet, and sex. A random forest regression model based on gene expression patterns was developed to predict acellular capillary density. Additionally, potential drugs that can attenuate early diabetic retinopathy were also identified.

Summary: This study provides a systematic review of the role and mechanism of DNA damage repair in germ cell development. It summarizes the causes, detection methods, and repair methods of DNA damage, as well as the mechanism of DNA damage repair. It also discusses the causes of abnormal DNA damage repair in germ cells, the effects of germ cell damage, and the applications of DNA damage repair in reproductive diseases and related surgical treatments.
