Interleukin-10 Producing Regulatory B Cells Transformed CD4+CD25- Into Tregs and Enhanced Regulatory T Cells Function in Human Leprosy

Regulatory B cells (Bregs) are known to exhibit their regulatory functions through interleukin-10 (IL-10) cytokine which suppress inflammation. There are only a few studies explaining the phenotype and functioning of these cells in contribution to host immunity in leprosy. Here, we evaluated the role of IL-10 producing Bregs in the pathogenesis of leprosy and assessed their immunoregulatory effects on Tregs and effector T cells. We found an increased frequency of Bregs and increased expression of their immune modulatory molecules (IL-10, FoxP3, and PDL-1) in leprosy patients. The potential immunoregulatory mechanism of Bregs was also investigated using MACS sorted Teff (CD4+CD25 ) and Treg (CD4+CD25+) cells were cocultured with Bregs to elucidate the effects of Bregs on effector T and regulatory T cells. Cell coculture results showed that purified Bregs cells from leprosy patients convert CD4(+)CD25(-) cells into CD4(+)CD25(+) cells. Cell coculture experiments also demonstrated that leprosy derived IL-10 producing Bregs enhance FoxP3 and PD-1 expression in Tregs and enhanced Tregs activity. Blocking of IL-10 receptor confirmed that IL-10 producing Breg has immunomodulatory effect on Tregs and effector T cells as effector T cells are not converted into Tregs and enhanced expression of FoxP3 and PD-1 was not observed on Tregs. Collectively, these findings demonstrate that IL-10 producing Breg cells play an important mechanism in controlling the immunopathogenesis of leprosy and have an immunomodulatory effect on Tregs and effector T cells. Our findings may pave way for novel targets of IL-10 producing Bregs for immunotherapy in leprosy patients.

Husk-like Zinc Oxide Nanoparticles Induce Apoptosis through ROS Generation in Epidermoid Carcinoma Cells: Effect of Incubation Period on Sol-Gel Synthesis and Anti-Cancerous Properties

Summary: This study effectively reports the influence of experimental incubation period on the sol-gel production of husk-like zinc oxide nanoparticles (ZNPs) and their anti-cancerous abilities. The surface morphology and diameter range of ZNPs were studied using SEM and TEM, respectively. The X-ray diffraction investigation showed that ZNPs had a pure wurtzite crystal structure. Prolonging the experimental incubation resulted in a relative drop in aspect ratio and a distinct blue-shift in the UV-visible spectrum. RBC lysis assay and MTT assay results indicated the innoxious nature and cytotoxicity of ZNPs against cancer cells. ZNPs may be useful in the treatment of cancer.

Targeting a transcription factor NF-?B by green tea catechins using in silico and in vitro studies in pancreatic cancer

Summary: This study aims to investigate the inhibitory mechanism of catechin derivatives (EC, EGC, ECG, and EGCG) on NF-kappa B in pancreatic cancer. In silico and in vitro studies were conducted to explore the binding pose, interacting residues, and molecular interactions. The results show that the catechin derivatives act as covalent inhibitors with the p65 subunit of NF-kappa B and interact with other residues through non-bonding interactions and hydrogen bonds. In addition, EGCG effectively reduces cell growth and proliferation, induces apoptosis, and inhibits NF-kappa B activity in pancreatic cancer cell lines, suggesting its potential as a therapeutic alternative for pancreatic cancer treatment.

Interlinking of diabetes mellitus and cancer: An overview

Summary: Cancer and diabetes mellitus are major causes of death and illness worldwide. Recent studies have shown that diabetes may promote cancer, although the exact mechanisms behind this association are largely unknown. This review aims to explore and understand the possible mechanisms linking diabetes and cancer. Hyperglycemia is a possible explanation for carcinogenesis in diabetic patients, as high glucose levels can aid in cancer cell proliferation. Chronic inflammation, another well-known factor in diabetes, may also play a role in the development of cancer. Additionally, the medications used to treat diabetes can either increase or reduce the risk of cancer. Insulin, a potent growth factor, can directly promote cell propagation and induce cancer, while hyperinsulinemia can increase the activity of growth factor-1. To improve cancer prognosis, individuals with diabetes should be screened for cancer at an early stage and receive appropriate treatment.

Among the investigated natural flavonoids, flavopiridol showed the highest potential as an inhibitor of the PI3K gamma pathway, providing new insights into rational drug discovery research for cancer therapy.
