IL-10 is identified as the critical factor for MDSCs therapy.

Summary: This study utilizes single-cell RNA sequencing and in vitro and in vivo models to explore the heterogeneity of extranodal natural killer/T-cell lymphoma (NKTCL). They identify a subset of malignant NK cells with EBV-encoded LMP1 that contribute to tumorigenesis and interact with immunocytes, impairing chemotaxis and exhibiting an immunosuppressive effect on T cells. High transcription of EBV-encoded genes and low infiltration of tumor-associated macrophages (TAMs) are favorable prognostic indicators for NKTCL. This study provides insights into the pathogenic mechanisms of NKTCL and potential therapeutic strategies.

Summary: Previous studies have shown that host immunity in paracoccidioidomycosis (PCM) is regulated by tolerogenic plasmacytoid dendritic cells, IDO-1, and Tregs. This study investigated the involvement of MDSCs in PCM and found that they played a role in immunosuppression. MDSCs were found to infiltrate the lungs during Paracoccidioides brasiliensis infection and their reduction resulted in a robust T-cell response and reduced disease severity. The study suggests that MDSCs could be a potential target for adjuvant therapy in PCM.

Summary: In this study, a soluble NKCAR was designed to trigger NK cell cytotoxicity by binding with the ligand MICA through the NKG2D receptor expressed on NK cells. The NKCAR showed bifunctional activity, recognizing both the CD20 antigen of target cells and the NKG2D receptor of NKL cells. The soluble NKCAR mediated the killing of CD20(+) Daudi cells in vitro, indicating its potential clinical application for killing target tumor cells.

Summary: This study investigated the therapeutic effects of IL-1 beta-primed ERCs in the attenuation of experimental colitis. The results showed that IL-1 beta-primed ERC treatment significantly reduced colonic damage and promoted the proliferation of anti-inflammatory cell populations in the spleen. Additionally, IL-1 beta pre-stimulation effectively downregulated DKK1 expression in ERCs, leading to enhanced therapeutic effects in colonic and splenic tissues.

Summary: The clinical benefits of immune checkpoint blockage therapy have been widely reported and it has been shown to enhance antitumor cytotoxic T cells. Neoantigens derived from tumor somatic mutations are believed to be ideal immune targets. Identifying neoantigens can help predict immunotherapy effectiveness and targeting them can be an innovative strategy for cancer treatment.

Summary: Natural killer T (NKT) cells and natural killer (NK) cells play important roles in the innate immune response to tumors and microorganisms by producing inflammatory cytokines and chemokines. The status and function of these cells in the tumor microenvironment (TME) have been investigated. Bacteria producing NKT cell ligands have been found to have anti-tumor effects, while tumor-derived lipids or metabolites may suppress NKT and NK cells. The balance between inhibitory and activating receptor signals determines the status and function of NKT and NK cells.
