Summary: AMPK, a key regulator of energy homeostasis, plays important roles in maintaining cardiac function under increased workload conditions. Deletion of AMPK genes in cardiomyocytes did not affect heart function under physiological conditions, but led to alterations in response to increased workload. AMPK deletion also resulted in decreased basal metabolic rate and locomotor activity, along with changes in cardiac mitochondrial function and bioenergetics.

Studies have shown that RSV infection leads to inhibition of AMPK activity, promoting downstream lipogenic effector activation and cellular lipid synthesis in HEp-2 cells.

Hyperglycemia induced similar changes in trophoblasts, and these changes were rescued by AMPK activation. AMPK activation upregulates trophoblast glucose uptake by stimulating GLUT3 translocation, which is beneficial for viability.

Furthermore, posttranscriptional mechanisms were proposed to be involved in the synthesis regulation of the heat shock protein 70-2 protein.

A study found that AMP-activated protein kinase (AMPK) plays a crucial role in maintaining energy metabolism and responding to insecticide treatment in N. lugens. Knockdown of NlAMPK alpha significantly enhanced the insecticidal efficiency of pymetrozine against N. lugens.

Summary: Skeletal muscle is an ideal target for lowering blood glucose through pharmacological interventions. Small molecule activators of AMPK that bind to the ADaM site can increase glucose uptake independently of AMPKg3 and when co-stimulated with AICAR, can further enhance muscle glucose uptake and AMPK activation. In vivo, PF739 treatment reduces blood glucose levels and selectively activates muscle AMPKg1 complexes.
