Herpes simplex virus type 1 (HSV-1) is a widespread neurotropic pathogen responsible for a range of clinical manifestations. Inflammatory cell infiltrate is a common feature of HSV-1 infections and has been implicated in neurodegeneration. Therefore, viral recognition by innate immune receptors (i.e., TLR2) and the subsequent inflammatory response are now deemed key players in HSV-1 pathogenesis. In this study we infected with HSV-1 the enteric nervous system (ENS) of wild-type (WT) and TLR2 knock-out (TLR2(ko)) mice to investigate whether and how TLR2 participates in HSV-1 induced neuromuscular dysfunction. Our findings demonstrated viral specific transcripts suggestive of abortive replication in the ENS of both WT and TLR2(ko) mice. Moreover, HSV-1 triggered TLR2-MyD88 depend signaling in myenteric neurons and induced structural and functional alterations of the ENS. Gastrointestinal dysmotility was, however, less pronounced in TLR2(ko) as compared with WT mice. Interesting, HSV-1 caused up-regulation of monocyte chemoattractant protein-1 (CCL2) and recruitment of CD11b(+) macrophages in the myenteric ganglia of WT but not TLR2(ko) mice. At the opposite, the myenteric plexuses of TLR2ko mice were surrounded by a dense infiltration of HSV-1 reactive CD3(+) CD8(+)INF gamma(+) lymphocytes. Indeed, depletion CD3(+) CD8(+) cells by means of administration of anti-CD8 monoclonal antibody reduced neuromuscular dysfunction in TLR2(ko) mice infected with HSV-1. During HSV-1 infection, the engagement of TLR2 mediates production of CCL2 in infected neurons and coordinates macrophage recruitment. Bearing in mind these observations, blockage of TLR2 signaling could provide novel therapeutic strategies to support protective and specific T-cell responses and to improve neuromuscular dysfunction in pathogen-mediated alterations of the ENS.

Molecular determinants of peritoneal dissemination in gastric adenocarcinoma

Valentina Mari, Valentina Angerilli, Giada Munari, Marco Scarpa, Quoc Riccardo Bao, Salvatore Pucciarelli, Matteo Fassan, Gaya Spolverato

Summary: This review outlines the molecular pathways involved in peritoneal metastatic dissemination in gastric cancer, highlighting the lack of comprehensive understanding of the complex molecular landscape of this disease. The integration of molecular profiling and histological evaluation is recommended for risk prediction assessment of peritoneal dissemination.

Tumor immune microenvironment in therapy-naive esophageal adenocarcinoma could predict the nodal status

Summary: This study found that immune marker expression, oncogene status, and intra- and peritumoral immune infiltrating cells are associated with lymph node metastasis in patients with esophageal adenocarcinoma. These results suggest that immune surveillance failure is the key driver of lymph node metastasis.

Microfluidic Strategies for Extracellular Vesicle Isolation: Towards Clinical Applications

Alessio Meggiolaro, Valentina Moccia, Paola Brun, Matteo Pierno, Giampaolo Mistura, Valentina Zappulli, Davide Ferraro

Summary: Extracellular vesicles (EVs) are receiving considerable attention in the biological and medical fields due to their role as natural carriers of genetic information and their potential as diagnostic and prognostic biomarkers in various diseases. However, conventional methods for EV isolation have limitations, and microfluidic approaches have been proposed as a solution. This review summarizes the most important microfluidic-based devices for EV isolation and their advantages and disadvantages compared to existing technology.

Collagen VI promotes recovery from colitis by inducing lymphangiogenesis and drainage of inflammatory cells

Sibilla Molon, Paola Brun, Melania Scarpa, Dario Bizzotto, Gaia Zuccolotto, Marco Scarpa, Matteo Fassan, Imerio Angriman, Antonio Rosato, Paola Braghetta, Ignazio Castagliuolo, Paolo Bonaldo

Summary: The study investigates the role of collagen VI in the development of intestinal inflammation and finds that it plays an important role in lymphatic vessel development and intestinal inflammation. The study also reveals that high collagen VI expression in the colon biopsies of CD patients is associated with increased surgical risk and disease recurrence.

Summary: This study evaluated the sex differences in the immune microenvironment of rectal cancer patients and found that male patients had more frequent oncogene mutations associated with lower expression of T-cell activation genes, while female patients had more Th1 cells and cytotoxic T cells in the healthy mucosa, suggesting a potentially better immune response to the tumor.

Summary: Plant cell cultures have emerged as a promising tool for producing active molecules, such as flavonols and anthocyanins, which have beneficial health properties. The synthesis of these molecules is finely regulated by specific transcription factors, and high expression levels can be achieved in tobacco cell lines. A single-column-based purification protocol was developed to produce a stable lyophile with antioxidant, anti-inflammatory, antibacterial, and wound-healing activities.

Summary: In this study, altered RNA editing in keratoconus (KC) was investigated. It was found that KC patients had significantly lower RNA editing levels, resulting in lower editing frequency and less edited bases. Differences in editing sites were observed in the chromosome 12 regions encoding Keratin type II cluster. Further investigation is needed to understand the functional implications.
