Summary: The study demonstrates the differences in site-specific glycosylation between virus-derived spikes, wild-type non-stabilized spikes, and recombinant engineered spike glycoproteins. Additionally, it shows that their unique cellular secretion pathways lead to distinct protein glycosylation and secretion patterns, including shedding of spike monomeric subunits for the non-stabilized wild-type spike, potentially impacting the resulting immune response and vaccine design.

Summary: This study updates potential targets and therapeutic drugs for SARS-CoV-2 from the perspective of the viral replication cycle, focusing on virus entry, proteases, and RdRp. Inhibitors for virus entry and RdRp are currently in clinical trials, while protease inhibitors require further in vivo and in vitro experiments to prove effectiveness against SARS-CoV-2.

Summary: This article summarizes how viruses alter major cellular metabolic pathways to support their replication needs, emphasizing the utilization of cellular signaling cascades and the changes in cellular nutrition and metabolism during viral infections. Studying these changes enhances understanding of fundamental metabolic regulation processes and offers potential for combatting viral diseases, metabolic disorders, and cancer.

Summary: Viral macrodomains are conserved in several virus families and have the potential to be targeted by drugs. These macrodomains remove posttranslational modifications on host or viral proteins to enhance replication. This study demonstrates the importance of poly-ADP-ribosylhydrolase (ARH) activity in efficient viral infection and replication.

Summary: This study reveals that CVB infection induces autophagy and increases the release of virus-laden EVs. Loss of TBK1 exacerbates CVB infection and increases the release of virus-laden EVs. This discovery sheds new light on the mechanisms involved in viral spread and EV propagation during acute enteroviral infection and highlights novel intracellular trafficking protein targets for antiviral therapy.

Summary: This study explores the evolution, key mutations, and features of the SARS-CoV-2 genome in the United States, and found many accumulating Single Nucleotide Variations (SNVs) in the virus genome throughout 2020 and early 2021. The results suggest that viral genomes are dynamically evolving over time, with periods of mutational bursts and unabated mutation accumulation. The high level of existing variation, especially in the Spike-encoding region, may become problematic during super-spreader events, driving rare mutations to prominence.

Summary: To study the function of the DPV pUL48 protein, researchers constructed a UL48-deleted mutant and a revertant virus using the red recombination system. The UL48-deleted mutant had reduced virus titers and efficiency of producing infectious virions, as well as smaller plaque sizes compared to the parental and revertant viruses. This study revealed the important role of UL48 as a late gene in viral replication and transmission, which can help in understanding the function of DPV pUL48 protein and preventing and controlling DPV.

Summary: Transposon-based insertional mutagenesis screens have shown that disruption of HCMV UL30 gene inhibits viral replication. Deletion of the canonical UL30 ORF reduces production of infectious virus, while comutation of methionines in the UL30 region and mutation of the TATA box lead to viral replication defects. The UL30-associated ORFs play an important role in HCMV infection and further research could provide insights for therapeutic interventions.
