Lactic Acid Bacteria Strains Exert Immunostimulatory Effect on H-pylori-Induced Dendritic Cells

The aim of this study was to find out if selected lactic acid bacteria (LAB) strains (antagonistic or nonantagonistic against H. pylori in vitro) would differ in their abilities to modulate the DCs maturation profiles reflected by their phenotype and cytokine expression patterns. Methods. Monocyte-derived DCs maturation was elicited by their direct exposure to the LAB strains of L. rhamnosus 900 or L. paracasei 915 (antagonistic and nonantagonistic to H. pylori, resp.), in the presence or absence of H. pylori strain cagA+. The DCs maturation profile was assessed on the basis of surface markers expression and cytokines production. Results. We observed that the LAB strains and the mixtures of LAB with H. pylori are able to induce mature DCs. At the same time, the L. paracasei 915 leads to high IL-10/IL-12p70 cytokine ratio, in contrast to L. rhamnosus 900. Conclusions. This study showed that the analyzed lactobacilli strains are more potent stimulators of DC maturation than H. pylori. Interestingly from the two chosen LAB strains the antagonistic to H. pylori-L. rhamnosus strain 900 has more proinflammatory and probably antibactericidal properties.

Summary: Dendritic cells play a crucial role in T cell alterations in primary hypertension. The study shows that the numbers and maturation markers of DCs in adolescent patients can reflect hypertension-mediated organ damage. Characteristics of DCs in hypertensive patients are associated with organ damage, with reduced tDC and pDC numbers, increased mDC subsets, and other changes.

Bladder Cancer Cells Exert Pleiotropic Effects on Human Adipose-Derived Stem Cells

Summary: Stem cell-based therapies are considered promising for treating bladder cancer. However, the delivery of cells producing paracrine factors may disrupt the microenvironment and potentially reactivate tumors. In this study, we investigated the interaction between adipose-derived stem cells (ASCs) and bladder cancer cells. We found that ASCs increased MCP-1 secretion by bladder cancer cells and altered their morphology. Additionally, soluble factors secreted by bladder cancer cells decreased ASC numbers and affected their behavior. Our results suggest that the interactions between ASCs and bladder cancer cells have a significant impact on the non-muscle-invasive urinary bladder.

Summary: Atherosclerosis is a leading cause of cardiovascular diseases and a major public health challenge. The mechanism of atherosclerosis is not fully understood, but alterations in lipid metabolism and inflammation play important roles. Recent research has shown significant effects of trained immunity on atherosclerotic plaque formation and development. Trained immunity is also a prominent factor in acute and chronic coronary syndrome.

Evaluation of Biological Activity of New 1,2,4-Triazole Derivatives Containing Propionic Acid Moiety

Summary: The aim of this research was to synthesize new 1,2,4-triazole derivatives containing propanoic acid moiety and investigate their anti-inflammatory, antibacterial, and anthelmintic activity. The structures of the derivatives were confirmed through various analysis methods. The compounds 3a, 3c, and 3e exhibited the most beneficial anti-inflammatory effects in cell culture experiments.

Summary: Viral infections and exposure to inhaled allergens, such as rhinovirus and house dust mite, are associated with asthma exacerbations. In this study, the authors found that rhinovirus-induced activation of the RIG-I inflammasome suppresses antiviral immunity, promotes inflammation, and aggravates subsequent infection with SARS-CoV-2, particularly after exposure to house dust mite. Understanding these mechanisms could lead to more effective treatment strategies for asthma and respiratory virus infections.

Alpha-Synuclein mRNA Level Found Dependent on L444P Variant in Carriers and Gaucher Disease Patients on Enzyme Replacement Therapy

Summary: Gaucher disease, caused by GBA1 gene variants, is characterized by hepatosplenomegaly, hematological abnormalities, and bone disease. GBA1 variants are also important risk factors for Parkinson's disease. A study investigated biomarkers for Gaucher disease and Parkinson's disease in patients receiving enzyme replacement therapy (ERT). The results showed elevated levels of alpha-synuclein mRNA in GD3 patients and L444P carriers, while GD1 patients, GBA1 carriers with unknown variants, and healthy controls had low levels of alpha-synuclein mRNA. The level of alpha-synuclein mRNA did not correlate with age in GD patients treated with ERT, except for L444P carriers.

Disparities in Cisplatin-Induced Cytotoxicity-A Meta-Analysis of Selected Cancer Cell Lines

Summary: Cisplatin, a widely used anticancer drug, shows unexpected diversity in cytotoxicity values in different tumor cell lines. After reviewing data from 2018 to 2022, we found large differences between studies using the same cell line, indicating experimental heterogeneity as the main cause. The available data is therefore diverse and not suitable as a reference, and researchers are advised to conduct preliminary testing before experiments.

Summary: Despite promising results in clinical trials, the use of immune checkpoint inhibitors (ICI) in acute myeloid leukaemia (AML) is limited. This study analysed the expression patterns of selected checkpoint proteins in AML patients and found that the expression of certain proteins was associated with the response to therapy and overall survival. Further validation through multicentre studies is needed.

The application of the natural killer cells, macrophages and dendritic cells in treating various types of cancer

Summary: Innate immune cells play an important role in tumor surveillance and clearance, and harnessing their function has proven to be an effective therapy for cancer treatment. Various immunotherapeutic strategies, including pharmacological agents, vaccines, cell transfer, and oncolytic virus-based therapy, are being investigated for anti-tumor activity. This review focuses on the preclinical and clinical applications of NK cell, macrophage, and DC immunotherapy in cancer treatment.
