Summary: The study reveals a crucial role for Notch2 in maintaining lung vascular endothelial cell quiescence and in PAH, with loss of Notch2 activating Notch1 and inducing proliferation and apoptosis resistance in human pulmonary artery endothelial cells. PAH patients exhibit reduced levels of endothelial Notch2 in pulmonary arteries, suggesting Notch2 as a fundamental driver of PAH pathogenesis.

Summary: We found that plasma circGSAP levels were decreased in IPAH patients and associated with poor outcomes. Overexpression of circGSAP improved survival and alleviated pulmonary vascular remodeling in MCT-PH rats. In vitro, circGSAP overexpression inhibited hypoxia-induced PMECs proliferation, migration, and increased mortality by absorbing miR-27a-3p. BMPR2 was identified as a target gene of miR-27a-3p and its silencing ameliorated the effect of the miR-27a-3p inhibitor on PMECs. The levels of BMPR2 were upregulated in circGSAP-overexpressed PMECs and lung tissues of MCT-PH rats.

Summary: This study reveals a novel mechanobiological mechanism underlying the dedifferentiation and proliferation of venous smooth muscle cells (SMCs) in neointimal hyperplasia. The expression of circSlc81/miR-20a-5p/Lamtor1 axis induced by arterial cyclic stretch may be a potential clinical target that attenuates neointimal hyperplasia in grafted vessels.

Summary: The study revealed that high expression of miR-27a in cholangiocarcinoma patients is associated with lymph node metastasis, TNM stages, and poorer overall survival. Overexpression of miR-27a promotes cell proliferation, migration, and invasion in CCA, while knockdown suppresses these processes. These findings suggest the potential utility of miR-27a in prognosis and progression of CCA.

Summary: Endothelial cells, which line blood vessels, are affected by biophysical cues and play an important role in vascular health and disease. This study investigated the impact of vessel stiffness on endothelial cells and found that cells cultured on softer substrates showed different cellular alignment and morphology compared to cells on stiffer substrates.

Summary: A T/C variant in miR-27a is associated with bipolar disorder by reducing its ability to target important neurodevelopmental genes. This study reveals the relationship between genetic variations of miRNA and schizophrenia or bipolar disorder, emphasizing the importance of miR-27a and its polymorphism in bipolar disorder.

Summary: Mechanical forces on cell-cell adhesion regulate barrier function of endothelial cells, with BRAF playing a crucial role in stress-induced responses. Actin cytoskeleton remodeling and intercellular gap formation are key factors influenced by external stresses, modulated by BRAF to potentially reverse decreased barrier function in blood vessels.

Summary: The quality of wool in agricultural animals is related to the development of hair follicles, which is influenced by hair follicle stem cells (HFSCs) and microRNA miR-27a. This study found that miR-27a inhibits the AKT/MTOR signaling pathway by targeting PIK3R3, thereby affecting the proliferation and apoptosis of HFSCs. These findings provide new insights into improving sheep wool quality.

Summary: A study found that the proliferation of trophoblasts in early-onset preeclampsia (ePE) is regulated by lncRNA MALAT1. The level of MALAT1 was decreased in placental tissue samples from ePE patients, and it was positively correlated with neonate birth-weight. Knockdown of MALAT1 inhibited trophoblast cell growth and migration, and promoted cell apoptosis. Moreover, MALAT1 reduction affected cell proliferation and growth through the regulation of miR-101-3p and VEGFA.

Summary: Our study reveals a critical role of microRNA-192-5p in mediating NOX-dependent DHFR deficiency and AAA formation, and inhibition of which effectively attenuates the development of AAA. Since the sequences of mouse and human microRNA-192-5p are identical, microRNA-192-5p inhibitors may offer a potential therapeutic option for AAA treatment.

Summary: Deletion of Ets1 gene leads to decreased migration of neural crest cells in mice, resulting in malalignment of cushions relative to the muscular ventricular septum similar to DORV defects in humans. Studies show that Ets1 gene disruption reduces migration speed and impairs cell-to-cell interactions in cardiac neural crest cells.

Summary: Studies have shown that combining folic acid with nifedipine has significant therapeutic effects on aortic aneurysms. Folic acid alone can effectively alleviate the development of aneurysms, while the combination with nifedipine further reduces the incidence of aneurysms and related complications. This innovative and powerful combination therapy can serve as a first-line oral medication for the treatment of aortic aneurysms.
