Summary: Pancreatic inflammation caused by bacterial endotoxin lipopolysaccharide (LPS) can alter cellular metabolism, autophagy, apoptosis, and proliferation, increasing the risks for organ toxicity and cancer. This study investigated the role of the antioxidant azadirachtin (AZD) in protecting pancreatic beta-cells from LPS-induced oxidative stress, showing that AZD induced autophagy signals for cell survival. Balancing the molecular communication between autophagy and apoptosis may be crucial for pancreatic cell survival and death under inflammatory and pathological conditions.

Summary: Lipids are an important energy source for insect reproduction, making them a target for pest management. The insecticide Azadirachtin (AZA) shows promise but its reproductive toxicity mechanism related to lipid metabolism is not well understood. This study used lipidomic and transcriptomic approaches to investigate the effects of AZA on lipid remodeling in the ovary of Spodoptera litura. The results revealed significant alterations in the content of 130 lipid subclasses, including changes in fatty acyl chain length and saturation degrees, as well as gene expression related to fatty acid biosynthesis and elongation. Western blot analysis also indicated that AZA induced insulin resistance-like phenotypes by inhibiting the insulin receptor substrate (IRS)/PI3K/AKT pathway. These findings provide insights into the lipid metabolism events and potential mechanisms involved in AZA exposure, and could serve as important references for future pest control.

Summary: iTRAQ-based comparative proteomic analysis revealed proteins and pathways involved in ovarian development and response to Azadirachtin exposure. Proteins related to proteasome and branched chain amino acids were essential for pupae-to-adult transition, while glycosylation-related pathway and proteins involved in vitamin B6 synthesis were necessary for ovary development under normal and AZA-exposure condition. Knockdown experiments confirmed the important roles of identified proteins in moth growth and ovary development.
