A retrospective study on EU harmonised classifications for carcinogenicity to guide future research

Summary: Under European Regulation (EC) No 1272/2008, chemicals can be classified as carcinogenic based on their ability to induce tumors, increase tumor incidence, malignancy, or shorten the time to tumor occurrence. Selecting the appropriate classification can be challenging, requiring consideration of hazard data and its relevance to humans. The European Chemical Agency's decisions on cancer classification are based on both the weight-of-evidence and strength-of-evidence of the particular activity.

Summary: Before registering and marketing new pharmaceutical, (agro)chemical or food ingredient products, manufacturers are required by law to generate data to ensure human safety. The selection of dose levels is crucial in designing studies to identify exposure levels that pose relevant hazards. As the science of predictive human exposure develops, there are exciting opportunities for more human-relevant approaches to dose level selection.

Reproductive and developmental evaluations of triclopyr acid, triclopyr butoxyethyl ester and triclopyr triethylamine salt in the rat

Summary: This paper presents the results of a rat two-generation study on triclopyr acid and provides a review of all available reproductive and developmental toxicity data from rat studies. The study found that developmental toxicity was only observed at the highest dose, which was likely due to maternal toxicity. Triclopyr was not found to be specifically toxic to reproduction or selectively toxic to the embryo, fetus, or neonate in the rat.

Developmental toxicity studies on triclopyr acid, triclopyr butoxyethyl ester and triclopyr triethylamine salt in the rabbit

Summary: Developmental toxicity studies have shown that triclopyr acid and its active-ingredient variants are not specifically toxic to rabbit embryos and fetuses, as post-implantation loss only occurred at doses causing maternal toxicity. However, T-BEE may be associated with increased post-implantation loss, but it was only observed in association with maternal toxicity.

Summary: The current agrochemical safety evaluation paradigm is outdated and does not address the challenges posed by population growth, food insecurity, and climate change. To support sustainable agriculture, the Health and Environmental Sciences Institute (HESI) has formed a technical committee consisting of international experts to establish a framework for modernizing the safety evaluation of crop-protection products. This framework aims to incorporate evolving science and meet the changing global and local needs and regulatory decisions.

Application of Defined Approaches for Skin Sensitization to Agrochemical Products

Summary: The skin sensitization potential of agrochemical formulations can be evaluated effectively using non-animal test methods, with the KeratinoSens assay showing the best performance in predicting in vivo hazard outcomes. Defined approaches (DAs) that integrate data from multiple non-animal methods are accepted for hazard classification and potency categorization. However, further research and data generation are needed to improve the accuracy and reliability of these methods.
