Summary: SCP has emerged as a novel intracellular delivery system with enhanced efficiency and serum tolerance. However, its application is limited due to cationic charge and non-specificity. Researchers have engineered an activatable SCP (ASCP) with pH-sensitive charge shielding sequence, protease cleavage site, and SCP. This system reduces non-specific binding and delivers various cargo into the cytosol both in vitro and in vivo. The ASCP fusion protein enables co-delivery of therapeutic cargos for optimal outcomes and paves the way for combination cancer therapies.

Summary: Simultaneous delivery of different functional drugs to tumor sites for chemotherapy remains challenging. In this study, nanogels formed by an engineered coiled-coil polypeptide PC(10)A were designed and prepared as a carrier for co-delivery of paclitaxel and doxorubicin. The results showed that PC(10)A/PTX/DOX nanogels had good biocompatibility, exhibited synergistic therapy effect of tumors, and had lower side effects compared with free PTX/DOX.

Summary: The development of nanotechnology has had a significant impact on biomedical and pharmaceutical fields, particularly in the design and formulation of stimuli-responsive nano-drug delivery systems. These systems, known as smart drug delivery systems, are able to respond to various external and internal stimuli and deliver drugs to targeted sites in a controlled manner. They offer improved patient compliance, treatment efficiency, and reduced side effects on healthy tissue. Polymeric nanogels with stimuli-responsive characteristics have shown great potential in biomedical, tissue engineering, and pharmaceutical applications due to their small size, biocompatibility, stimuli-triggered drug deliverability, high payload capacity, and tailored functionality.

The material showed excellent selectivity and high sensitivity, allowing for the determination of the biomarker in saliva and urine. Additionally, a convenient point-of-care testing method using fluorescent test paper and a smartphone was established for the initial diagnosis of anthrax.
