Biogeographic studies are important for understanding the natural history of faunas. To comprehend the geographical patterns of genetic variation in anurans in Taiwan, we investigated the genetic structure of Sylvirana latouchii (Anura, Ranidae) from 31 populations by using mitochondrial DNA (mtDNA) cytochrome b sequences. A neighbor-joining tree of 38 haplotypes revealed three major divergences in Taiwanese S. latouchii: the northern, western, and eastern-and-southern clades. Each clade was restricted to a single geographical district and showed obvious differentiation. The patterns of geographical divergence in this species reflect common historical events experienced by other native animals distributed in Taiwan. The order of divergence times between clades was inferred using a molecular clock test. The population relationship of S. latouchii between Taiwan and mainland China is discussed. Further study employing more populations of S. latouchii from mainland China is necessary to clarify the original geographical patterns and migratory history of this species.

Population genetic structure of the insular Ryukyu flying fox Pteropus dasymallus

Summary: Small isolated populations, such as the insular Ryukyu flying fox, are vulnerable to negative impacts of genetic drift and stochastic events. Genetic management of separate island subgroups is crucial for conservation efforts to prevent further declines in genetic diversity. Mitochondrial and microsatellite DNA analysis revealed significant genetic differentiation among island populations of P. dasymallus, likely influenced by historical colonization and oceanic barriers.

Hypothermal stress-induced salinity-dependent oxidative stress and apoptosis in the livers of euryhaline milkfish, Chanos chanos

Chia-Hao Chang, Yu-Chun Wang, Tsung-Han Lee

Summary: This study found that under low temperature, freshwater milkfish exhibit poorer cold tolerance and higher oxidative stress compared to seawater milkfish, leading to apoptosis in hepatocytes. Additionally, during low-temperature acclimation, more apoptosis and impaired liver functions were observed in freshwater milkfish compared to seawater milkfish.

Time-course changes in the regulation of ions and amino acids in the hard clam Meretrix lusoria upon lower salinity challenge

Chia-Hao Lin, Po-Ling Yeh, Tsung-Han Lee

Summary: This study investigated ion and amino acid regulation in the gill and mantle of the hard clam Meretrix lusoria. Results showed significant reductions in osmolality, Na+, and Cl- concentrations in the hemolymph of clams transferred to a lower salinity environment. Specific activities of NKA were unaffected, but there was a decline in FAA contents, with taurine identified as the dominant FAA. Additionally, mRNA expression and protein abundance of TAUT were altered in response to reduced taurine content in the gill and mantle of hard clams during acclimation.

Rapid response of osmotic stress transcription factor 1 (OSTF1) expression to salinity challenge in gills of marine euryhaline milkfish (Chanos chanos)

Yu-Ting Lin, Tsung-Han Lee

Summary: This study investigated changes in the expression levels of OSTF1, cortisol levels, and their correlation in the marine inhabitant milkfish. The results showed that ostf1b was the primary gene responding to hypertonic stress, and cortisol concentration increased after transferring milkfish from freshwater to seawater. Additionally, cortisol injection increased the expression of ostf1a and ostf1b.

Genetic diversity and structure of Eurasian otters on Kinmen Island

Nian-Hong Jang-Liaw, Yu-Chun Tan, Chia-Jui Chang, Chung-Hao Juan, Hsuan-Yi Hou, Li-Wai Chung, Huan-Song Cao, Daisuke Waku, Shih-Wei Chang, Ling-Ling Lee

Summary: In this study, the genetic diversity and structure of wild Eurasian otters (Lutra lutra) on Kinmen Island, China were analyzed using high-quality DNA samples from 40 individuals. Mitochondrial DNA sequencing revealed no nucleotide diversity among the samples. However, genotyping at microsatellite loci showed no deviation from Hardy-Weinberg equilibrium and no inbreeding depression. Phylogenetic analysis indicated that the Kinmen otter population is most genetically similar to a now-extinct population in Taiwan and forms a monophyletic group with southern Chinese populations. These findings can inform conservation programs for Eurasian otters in the Kinmen region.

Glucocorticoid Receptor Mediates Cortisol Regulation of Glycogen Metabolism in Gills of the Euryhaline Tilapia (Oreochromis mossambicus)

Chien-Yu Wu, Tsung-Han Lee, Deng-Yu Tseng

Summary: In this study, the effects of cortisol on the regulation of glycogen metabolism biomarkers in the gills of tilapia were investigated. It was found that cortisol regulated glycogen metabolism by promoting the expression of glycogen phosphorylase mRNA in the gills. These findings suggest that cortisol directly acts on glycogen-rich cells in the gills and regulates glycogen metabolism.

Salinity effects on expression and localization of aquaporin 3 in gills of the euryhaline milkfish (Chanos chanos)

Yu-Ting Lin, Shao-Ying Wu, Tsung-Han Lee

Summary: This study investigated the expression and localization of AQP3 in the gills of milkfish, contributing to our understanding of the physiological role and localization of AQP3 in fish. The results showed that the abundance of AQP3 protein significantly decreased 3 hours after transferring milkfish from fresh water to seawater, but remained unchanged within 7 days after hypotonic stress. Additionally, double immunofluorescence staining revealed the colocalization of AQP3 and Na+/K+ ATPase at the basolateral membrane of ionocytes in milkfish gills. These findings suggest that milkfish have a strong osmoregulation ability under acute osmotic stress due to the rapid changes in the gene and protein expression of AQP3 in their gills.
