The role of proteomics in progressing insights into plant secondary metabolism

The development of omics has enabled the genome-wide exploration of all kinds of biological processes at the molecular level. Almost every field of plant biology has been analyzed at the genomic, transcriptomic and proteomic level. Here we focus on the particular contribution that proteomic technologies have made in progressing knowledge and characterising plant secondary metabolism (SM) pathways since early expectations were created 15 years ago. We analyzed how three major issues in the proteomic analysis of plant SM have been implemented in various research studies. These issues are: (i) the selection of a suitable plant material rich in secondary metabolites of interest, such as specialized tissues and organs, and in vitro cell cultures; (ii) the proteomic strategy to access target proteins, either a comprehensive or a differential analysis; (iii) the proteomic approach, represented by the hypothesis-free discovery proteomics and the hypothesis-driven targeted proteomics. We also examine to what extent the most-advanced technologies have been incorporated into proteomic research in plant SM and highlight some cutting edge techniques that would strongly benefit the progress made in this field.

Proteome changes in pepper (Capsicum annuum L.) leaves induced by the green peach aphid (Myzus persicae Sulzer)

Summary: Aphid attacks trigger plant defense responses and lead to changes in the proteome of pepper leaves, with low density infestation resulting in a smaller number of differentially expressed proteins compared to high density infestation. The altered proteins are involved in various aspects of plant primary metabolism, with photosynthesis being the most affected process.

Rational Design of Resveratrol O-methyltransferase for the Production of Pinostilbene

Summary: This study reports the engineering of a resveratrol OMT from Vitis vinifera with high catalytic efficiency in producing pterostilbene. Through point mutations and variant design, the native substrate selectivity of VvROMT was successfully altered, indicating its potential for the tailor-made production of stilbenes.

Summary: Stilbenes, such as resveratrol and its derivatives, have interesting properties but limited by their poor aqueous solubility and low bioavailability. Cyclodextrins play a significant role in stilbene chemistry, forming nanomolecular inclusion complexes for drug delivery and synthesis of derivatives. Plant cell systems offer a feasible way for large-scale production of stilbenes, requiring elicitor compounds like cyclodextrins for biosynthesis induction.

Summary: The study found that under CD induction conditions, PPCK was down-regulated, stilbene production increased, and PPC activity was lost in both cell cultures and leaves. Additionally, STS transcripts were co-induced with MYB14 and WRKY24 in both tissues. These genes had not been previously reported to respond to CD in grape leaves.

Summary: This study presents the characterization and optimization of t-R production in grape cell cultures. Factors such as growth phase, elicitor concentration, biomass, order of addition, and cell conditions were investigated. The optimal conditions for t-R production were determined, and the results can guide the upscaling of the bioproduction system.

MITIGATION OF CADMIUM CONCENTRATION IN COCOA KERNELS

Summary: This study evaluated the use of pectintranseliminase (PTE) to mitigate cadmium concentration in cocoa beans through a fast fermentation process. The results showed that the application of PTE significantly reduced the cadmium content in cocoa beans, leading to improved physical and sensory quality of the beans.

Summary: The objective of this research was to evaluate the vegetative development of cuttings of tropical Eucalyptus clones with auxin application. The results showed that clone EC166 had better response to 1500 mg kg(-1) AIB, while the number of roots showed better response to 1000 mg kg(-1) ANA.
