Summary: This study compared the efficiency of AsCas12a nuclease from Acidaminococcus sp. with SpCas9 from Streptococcus pyogenes for functional genomics in Schistosoma mansoni. AsCas12a was more efficient in gene knockout, while SpCas9 was more precise in transgene insertion. Both nucleases had similar phenotypic impact on transcription and expression of omega-1.

Schistosoma mansoni α-N-acetylgalactosaminidase (SmNAGAL) regulates coordinated parasite movement and egg production

Summary: Alpha-galactosidase (α-GAL) and alpha-N-acetylgalactosaminidase (α-NAGAL) are two glycosyl hydrolases responsible for maintaining cellular homeostasis by regulating glycan substrates on proteins and lipids. The parasitic blood fluke Schistosoma mansoni also contains functionally important α-GAL and α-NAGAL proteins, and inhibiting their activities could lead to the development of novel chemotherapeutics. The study identified that Smp_089290 is the only S. mansoni protein containing the necessary amino acids for α-GAL/α-NAGAL substrate cleavage. Inhibition of α-NAGAL activity in the adult worms significantly reduced their motility and egg production. This study contributes to understanding the glycan processing and modification machinery of schistosomes and suggests that targeting α-NAGAL may be a potential strategy for controlling schistosomiasis.

Exploring the role of macrophages in determining the pathogenesis of liver fluke infection

Summary: This review investigates the differential activation of macrophages during infection with food-borne trematodes and its impact on the pathological outcome of infection. Infection with Fasciola hepatica suppresses the activation of M1 macrophages, while infection with Opisthorchis viverrini and Clonorchis sinensis promotes the predominance of M1 macrophages in the bile ducts. The activation of M1 macrophages contributes to mutagenesis and tumor formation, while the induction of M2 macrophages promotes the proliferation of mutated cells and the expansion of cancerous tissue.

Knockout of liver fluke granulin, Ov-grn-1, impedes malignant transformation during chronic infection with Opisthorchis viverrini

Summary: Infection with the food-borne liver fluke Opisthorchis viverrini is the main risk factor for cholangiocarcinoma in the Mekong Basin countries. This study used a gene-edited liver fluke model to investigate the role of the fluke granulin-like growth factor Ov-GRN-1 in malignancy. The results showed that knocking out the Ov-GRN-1 gene reduced hepatobiliary tract disease and the development of high-grade cholangiocarcinoma in infected hamsters.

Summary: We have discovered a peptide called FhHDM-1 in the secretions of the liver fluke, which can regulate inflammation by reprogramming macrophage metabolism. Specifically, FhHDM-1 switches macrophage metabolism to rely on oxidative phosphorylation fueled by fatty acids and supported by the induction of glutaminolysis. This metabolic switch leads to a decrease in TNF and IL-6 production, indicating the potential of FhHDM-1 in preventing inflammation.

Exploring Evolutionary Relationships within Neodermata Using Putative Orthologous Groups of Proteins, with Emphasis on Peptidases

Summary: This study examined the phylogenetic relationships within Neodermata using putative orthologous groups of proteins (OGPs) from 11 species of Monogenea, Trematoda, and Cestoda. The analysis showed that monopisthocotylean monogeneans are sister-group to cestodes, and polyopisthocotylean monogeneans are sister-group to trematodes. However, the sister-group relationship between Monopisthocotylea and Cestoda was not statistically supported. The study also revealed a cluster formed by polyopisthocotyleans and trematodes.

Summary: Chronic human liver fluke infections can cause liver and biliary diseases, including the development of cholangiocarcinoma. We have developed an improved lateral flow immunochromatographic test kit that uses whole-blood samples to diagnose opisthorchiasis and clonorchiasis. The diagnostic values of the kit have been evaluated and it shows high accuracy.

How do parasitic worms prevent diabetes? An exploration of their influence on macrophage and beta-cell crosstalk

Summary: Diabetes is a rapidly growing chronic disease globally, with two distinct subtypes characterized by autoimmune destruction of pancreatic beta cells or metabolic insufficiency. Macrophages play a key role in the pathogenesis of both subtypes, influencing inflammation and beta-cell function. Studies have shown positive results in using parasitic worms and their products to modulate immune responses and promote beta-cell function and survival.
