Summary: The clinical successes of immune checkpoint blockade have led to efforts in activating T cell-mediated responses against cancer, with particular focus on the PVR family of inhibitory receptors. This study assessed the binding interactions and molecular assemblies of these receptors and ligands in vitro, and determined the crystal structure of the anti-TIGIT monoclonal antibody BMS-986207 in complex with TIGIT. The findings suggest that the PVR family members, including TIGIT and CD155, engage in interactions with relatively weak affinity, indicating that immune suppression is likely elicited through increased surface expression rather than inherent differences in affinity.

Summary: By controlling the symmetry and number of supramolecular interactions on protein surfaces, nonspecific interactions between proteins can be induced to form various nanoscale assemblies, including coiling nanowires, nanotubes, and nanosheets, without manipulating the protein's native surfaces. These nanoscale assemblies can spontaneously evolve into more ordered architectures, crystals. These findings reveal a transformation mechanism of protein self-assembly that has not been exploited before and may provide an approach to generate complex and transformable biomacromolecular self-assemblies.

Summary: Fabry disease is caused by a deficiency of α-galactosidase A (GLA), resulting in the accumulation of glycosphingolipids. Directed evolution was used to generate more stable GLA variants, such as GLAv05 and GLAv09, which showed improved stability, enzyme activity, and pharmacokinetics. These findings hold promise for more effective treatments of lysosomal storage diseases.
