Summary: Matrix mechanics, especially substrate stiffness, regulate cell behaviors such as viability, communication, migration, and differentiation. This study investigates the effects of substrate stiffness on the states of neutrophils (Neus) using a three-dimensional (3D) culture system made of hydrogels. It is found that a stiffer matrix promotes Neus to shift to an anti-inflammatory phenotype (N2) through the activation of the JAK1/STAT3 pathway. These findings provide insights into fabricating engineered tissue mimics and offer more treatment options for ischemic diseases and bone defects.

Loss of EPN3 weakened proliferation, migration, and invasion of NSCLC cells, induced apoptosis, and repressed epithelial-mesenchymal transition. EPN3 acted as an oncogene in NSCLC by activating the JAK1/2-STAT3 pathway.
