An effective antidote for paraquat poisonings: The treatment with lysine acetylsalicylate

Sodium salicylate (NaSAL) has been shown to have a multifactorial protection mechanism against paraquat (PQ)-induced toxicity, due to its ability to modulate inflammatory signalling systems, to prevent oxidative stress and to its capacity to chelate PQ. Considering that currently there is no pharmaceutical formulation available for parenteral administration of NaSAL, the aim of the present study was to evaluate the antidotal feasibility of a salicylate prodrug, lysine acetylsalicylate (LAS), accessible for parenteral administrations. PQ was administered to Wistar rats by gavage (125 mg/kg of PQ ion) and the treatment was performed intraperitoneally with different doses (100, 200 and 400 mg/kg of body weight) of LAS. Survival rate was followed during 30 days and living animals at this endpoint were sacrificed for lung, kidney, liver, jejune and heart histological analysis. it was shown, that the salicylate prodrug, LAS, available in a large number of hospitals, is also effective in the treatment of PQ intoxications. From all tested LAS doses, 200 mg/kg assured animal's full survival. Comparatively to 60% of mortality observed in PQ only exposed animals, the lethality was higher (80%) in the group that received 400 mg/kg of LAS 2 h after PQ administration. The dose of 100 mg/kg of LAS showed only a modest protection (60% of survival). Collagen deposition was observed by histological analysis in survived animals of all experimental groups, being less pronounced in animals receiving 200 mg/kg of LAS, reinforcing the importance of this dose against tissue damage induced by PQ. The results allow us to suggest that LAS should be considered in the hospital treatment of PQ poisonings. (C) 2008 Elsevier Ireland Ltd. All rights reserved.

Autophagy (but not metabolism) is a key event in mitoxantrone-induced cytotoxicity in differentiated AC16 cardiac cells

Summary: This study compares the cardiotoxicity of MTX and its metabolite NAPHT in an in vitro cardiac model and explores the role of metabolism in the cardiotoxic effects. The results show that MTX causes concentration-dependent cytotoxicity, affecting mitochondrial function and lysosome uptake, while NAPHT only exhibits concentration-dependent cytotoxicity. Inhibition or induction of metabolism has minimal impact on MTX-induced cytotoxicity, suggesting that metabolism is not a major contributor to MTX toxicity. However, autophagy is involved in MTX-induced cytotoxicity, potentially through the involvement of p62/LC3-II.

Is competitive swimming training a risk factor for osteoporosis? A systematic review of the literature and quality of evidence

Summary: This study analyzed the effect of swimming on bone mass, cortical geometry, and trabecular microarchitecture. The results showed that swimmers have lower bone mass compared to athletes engaged in high-impact sports, but similar to those engaged in moderate- and non-impact sports as well as non-athletes. Swimmers' cortical geometry and trabecular microarchitecture were also found to be similar to those of athletes and non-athletes. However, the evidence was of poor quality.

Protein Turnover in Skeletal Muscle: Looking at Molecular Regulation towards an Active Lifestyle

Summary: Skeletal muscle is a highly adaptable tissue that can change its mass and function in response to various stimuli. Protein synthesis and breakdown, regulated by multiple signaling pathways, play a crucial role in skeletal muscle mass. Different signaling pathways, such as Akt/mTOR signaling, ubiquitin-proteasome pathway, and autophagy, contribute differently to protein turnover and muscle mass depending on the condition and type of muscle. PGC-1α, by modulating mitochondria biogenesis, influences the cell's bioenergetic status and protein turnover. This review aims to provide an updated understanding of the molecular pathways involved in protein turnover regulation in skeletal muscle, with a focus on how lifestyle interventions, particularly exercise training, modulate various muscle proteins. An in-depth analysis of the anabolic effects of exercise programs will pave the way for personalized management of muscle wasting conditions.

Study of the potential toxicity of adrenaline to neurons, using the SH-SY5Y human cellular model

Summary: Prolonged overexposure to catecholamines causes toxicity due to continuous adrenoceptor stimulation, autoxidation, and reactive pro-oxidant species formation. This study investigated the contribution of oxidative stress in adrenaline-induced neurotoxicity using non-differentiated SH-SY5Y cells. Results showed that Adrenaline caused concentration- and time-dependent cytotoxicity in the cells, and N-acetyl-cysteine prevented the toxicity while the antioxidant Tiron was non-protective. Further analysis is needed to understand the involvement of these processes in catecholamine-induced peripheral neuropathy.

Assessment of the Permeability of 3,4-Methylenedioxypyrovalerone (MDPV) across the Caco-2 Monolayer for Estimation of Intestinal Absorption and Enantioselectivity

Summary: This study presents the first determination of the permeability of MDPV across an intestinal epithelial cell monolayer. The enantiomers of MDPV were found to have high permeability across the Caco-2 monolayer. Enantioselectivity and efflux through a facilitated diffusion mechanism were also observed.

Red-shifted and pH-responsive imidazole-based azo dyes with potent antimicrobial activity

Summary: This article describes a new route to obtain 2-aminoimidazole azo dyes with a unique substituent pattern in the heteroaryl unit that exhibit halochromic properties, changing from magenta to deep blue. The dyes also demonstrate potent antimicrobial properties against infectious yeasts. Cytotoxicity was not detected at concentrations lower than 16 μg mL(-1).
