Summary: Researchers have successfully generated skeletal muscle myoblasts from human induced pluripotent stem cells using an efficient and reliable transgene-free protocol. These myoblasts can be stably propagated and have the ability to differentiate into myotubes and regenerate reserve cells. In addition, they exhibit stem cell-like properties when engrafted into mouse muscle and are responsive to signals in the host muscle. This study demonstrates the utility of these myoblasts for investigating human myogenesis and disease pathogenesis, as well as for developing muscle stem cell therapeutics.

Summary: Cell replacement therapies using human embryonic stem cell and induced pluripotent stem-derived retinal ganglion cells (hRGCs) hold promise in achieving functional recovery in neurodegenerative optic neuropathies diseases. However, previous transplantation studies have shown limited success in terms of cell survival and integration in the host retina. In this study, a transplantation assay involving hRGCs and mouse retina explants was designed to overcome physical barriers and evaluate strategies for optimizing integration of donor cells. Results showed that supplementing cultures with brain derived neurotrophic factor (BDNF) and forskolin improved hRGC integrity, neurite outgrowth, and invasion of mouse explants.

Summary: This study investigated the impact of Wnt/β-catenin signaling activation on human limbal epithelial stem cells (hLESCs) using the GSK-3 inhibitor LY2090314. The results showed that activation of the Wnt/β-catenin signaling pathway led to downregulation of stemness genes and upregulation of differentiation genes, as well as reduced cell proliferation and stem cell maintenance capability.

Summary: This study evaluates the efficacy of in situ AAV-mediated gene delivery into the human corneal limbal region via targeted sub-limbal injection technique. The results demonstrate the feasibility and efficiency of the sub-limbal injection technique in specifically targeting cells in the human limbus. This technique may provide a feasible mode of genetic therapy for corneal disorders with an epithelial etiology.

Summary: The study demonstrates that the use of a designed peptide tag (SP)(20) can significantly increase the secretion yield of EPO and promote its extracellular secretion in plant cells. Additionally, the use of the reporter protein GFP allows for easy monitoring of protein expression. The (SP)(20)-tagged EPOs exhibit activity in promoting cell proliferation and hematopoietic cell differentiation.

Summary: Skeletal muscle stem and progenitor cells have the potential for personalized therapies, but their engraftment efficiency is low. This study shows that competition with mouse satellite cells hinders the engraftment of skeletal muscle progenitor cells into stem cell niches, but a mouse model with satellite cell ablation promotes their engraftment and regeneration. The expression of human regenerating myofibres is crucial for supporting skeletal muscle progenitor cells.

Summary: Different experimental strategies for ex vivo expansion of human hematopoietic stem cells (HSCs) have been developed, however, the genomic function of expanded HSCs under different culture systems remains unclear. This study compared gene expression profiles of HSCs in serum and serum-free culture systems, identifying differentially expressed genes enriched in the TNF-regulated inflammatory pathway in the FBS culture system. The ex vivo expansion of HSCs using the FBS culture system induces an inflammatory response and high CD38 expression, providing new insights into genomic functions of HSCs under different expanded cultures.

Summary: This study investigated the potential of human term placenta-derived amniotic membrane MSCs (hAMSC) to support the ex vivo expansion of HSC and progenitor cells. The results suggest that hAMSCs are a valuable alternative to BM-MSC for enhancing the proliferation and maintaining stemness of CB-HSC, and can improve the colony forming unit ability and long-term culture initiating cells ability, addressing the need for high HSC content in CB units available for transplantation.
