To compare the effects of alpha-ketoglutarate (alpha-KG) and melatonin on 24-h rhythmicity of oxidative stress in N-nitrosodiethylamine (NDEA)-injected Wistar male rats, melatonin (5 mg/kg i.p.) or alpha-KG (2 g/kg through an intragastric tube) was given daily for 20 weeks. In blood collected at 6 time points during a 24-h period, serum activity of aspartate transaminase (AST) and alanine transaminase (ALT) and the levels of alpha-fetoprotein (alpha-FP) were measured as markers of liver function. To assess lipid peroxidation and the antioxidant status, plasma levels of thiobarbituric acid reactive substances (TBARS) and of reduced glutathione (GSH) were measured, together with the activity of erythrocyte superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) and glutathione S-transferase (GST). NDEA augmented mesor and amplitude of rhythms in AST and ALT activity and plasma alpha-FP levels and mesor values of plasma TBARS, while decreasing mesor values of plasma GSH and erythrocyte SOD, CAT, GPx and GST. Acrophases were delayed by NDEA in all cases except for alpha-FP rhythm, which became phase-advanced. Co-administration of melatonin or alpha-KG partially counteracted the effects of NDEA. Melatonin decreased mesor of plasma TBARS and augmented mesor of SOD activity. The results indicate that melatonin and alpha-KG are effective in protecting from NDEA-induced perturbation of 24-h rhythms in oxidative stress. Melatonin augmented antioxidant defense in rats.

Inevitable interlinks between biological clock and metabolism in mammals

Perumalip Subramanian, Jaime Jacqueline Jayapalan, Puteri Shafinaz Abdul-Rahman

Summary: The perception of energy homeostasis involves the regulation between biological clock and cell metabolism, where the temporal regulation of metabolism, physiology, and behavior in mammals is coordinated by a complex system of cellular clocks synchronized by a master clock in the hypothalamus. Disruption of circadian rhythms, as seen in shift-workers, can lead to negative mental and metabolic disturbances, highlighting the close relationship between metabolic processes and the biological clock. Further research is needed to fully understand the functional connections between circadian rhythms and metabolism at different levels from molecular to organismic.

Timing is everything: Circadian rhythms and their role in the control of sleep

Seithikurippu R. Pandi-Perumal, Daniel P. Cardinali, Nevin F. W. Zaki, Ramanujam Karthikeyan, David Warren Spence, Russel J. Reiter, Gregory M. Brown

Summary: Sleep and the circadian clock are interconnected and vital to human health and well-being. The suprachiasmatic nucleus controls the sleep-wake cycle and homeostatic activities. Melatonin, as a part of the endogenous circadian clock, plays a critical role in sleep regulation. Light acts as a signal for sleep/wake control by affecting melatonin secretion. Clock genes serve as molecular timekeepers for maintaining the 24-hour cycle. The significance of these processes has been officially recognized in modern sleep and behavioral disorder diagnostic manuals. Timing in the brain is crucial for optimal brain physiology.

Molecular mechanisms interlinking biological clock and diabetes mellitus: Effective tools for better management

Chandrasekaran Sankaranarayanan, Perumal Subramanian

Summary: This article summarizes the link between circadian disturbances and diabetes mellitus, and discusses how these associations can be utilized for effective management of the disease. The study found that misalignment between peripheral clocks and the central oscillator alters the secretion of insulin, incretins, adipokines, and soluble factors, resulting in metabolic derangement and chronic hyperglycemia. Therefore, managing circadian health can restore glucose homeostasis, and circadian clock modifiers may serve as potential therapeutic agents for treating diabetes mellitus.

Summary: Nomophobia, a recent disorder receiving considerable attention, has been extensively studied since it was introduced in 2008. Most of the research focuses on determining its prevalence in different populations and its association with other mental and physical health issues. However, significant data on the treatment and prevention of nomophobia are lacking.

Summary: In this study, we examined the immediate impact of a social media outage on nomophobia and associated symptoms using a longitudinal cohort design. The results showed that symptoms of nomophobia increased significantly during the social media outage, and baseline insomnia scores predicted a surge in the global scores of nomophobia symptoms.
