Summary: In this study, the potential molecular mechanism underlying radiation-induced heart disease (RIHD) was investigated using RNA-sequencing (RNA-seq) and bioinformatics methods. A rat model of RIHD was established and differentially expressed circRNAs, miRNAs, and mRNAs were identified. Functional and signaling pathway analyses revealed that the differentially expressed mRNAs were most enriched in pathways related to endothelial function and vascular pathological processes. A candidate competitive endogenous RNA (ceRNA) network involving circRNA-miRNA-mRNA was established, which was further validated using qRT-PCR.

Summary: An integrative analysis of differentially expressed genes (DEGs) and miRNAs from ICC onset and adjacent normal tissues was conducted to explore the regulatory roles of miRNA-mRNA interaction. A total of 1018 DEGs and 39 miRNAs were found to be likely involved in ICC pathogenesis, indicating alterations in cell metabolism during ICC development. The network analysis revealed that 30 DEGs were regulated by 16 differentially expressed miRNAs. The identified DEGs and miRNAs may serve as biomarkers for ICC, highlighting their potential importance in ICC pathogenesis. This study provides a solid foundation for understanding the regulatory mechanism of miRNAs and mRNAs in ICC pathogenesis.

Summary: In this study, a computational framework was proposed to infer mRNA stability of genes using RNA-seq data and identify stQTLs. The results revealed that stQTLs were enriched in the CDS and 3'UTR regions, while eQTLs were enriched in multiple regions. Additionally, stQTLs were more likely to overlap with RNA binding protein and microRNA binding sites. This study demonstrates that simultaneous identification of stQTLs and eQTLs provides more mechanistic insight into the association between genetic variants and gene expression levels.
