Comparative proteomic analysis of pathogenic and non-pathogenic strains from the swine pathogen Mycoplasma hyopneumoniae

Background: Mycoplasma hyopneumoniae is a highly infectious swine pathogen and is the causative agent of enzootic pneumonia (EP). Following the previous report of a proteomic survey of the pathogenic 7448 strain of swine pathogen, Mycoplasma hyopneumoniae, we performed comparative protein profiling of three M. hyopneumoniae strains, namely the non-pathogenic J strain and the two pathogenic strains 7448 and 7422. Results: In 2DE comparisons, we were able to identify differences in expression levels for 67 proteins, including the overexpression of some cytoadherence-related proteins only in the pathogenic strains. 2DE immunoblot analyses allowed the identification of differential proteolytic cleavage patterns of the P97 adhesin in the three strains. For more comprehensive protein profiling, an LC-MS/MS strategy was used. Overall, 35% of the M. hyopneumoniae genome coding capacity was covered. Partially overlapping profiles of identified proteins were observed in the strains with 81 proteins identified only in one strain and 54 proteins identified in two strains. Abundance analysis of proteins detected in more than one strain demonstrates the relative overexpression of 64 proteins, including the P97 adhesin in the pathogenic strains. Conclusions: Our results indicate the physiological differences between the non-pathogenic strain, with its non-infective proliferate lifestyle, and the pathogenic strains, with its constitutive expression of adhesins, which would render the bacterium competent for adhesion and infection prior to host contact.

Expression and distribution of glutathione transferases in protoscoleces of Echinococcus granulosus sensu lato

Summary: The study revealed the important roles of GSTs in E. granulosus s.l. biology, particularly in parasite establishment and detoxification processes. EgGST1 was found to be associated with detoxification, with increased expression in the parenchyma of pre-adult protoscoleces (PSCs); EgGST2, located on the tegument of PSCs, showed increased expression post-anthelmintic treatment, potentially related to immunoregulation and chemotherapy resistance; EgGST3, related to the Omega class, exhibited distribution throughout the PSC body and increased expression in response to oxidative stress.

Summary: This study investigated the protein content of the hydatid fluid compartment in E. granulosus and E. ortleppi pulmonary bovine cysts and compared their molecular arsenal at the host-parasite interface. The results showed a predominance of parasite proteins compared to host proteins in the hydatid fluid samples. Functional analysis revealed important molecular functions and processes that are active in pulmonary cystic echinococcosis.

Are the Bacteria and Their Metabolites Contributing for Gut Inflammation on GSD-Ia Patients?

Summary: In this study, the fecal metabolites of GSD patients were measured and compared to healthy controls. The results showed higher levels of succinic acid in GSD patients, while no significant differences were observed for other metabolites. Fecal pH was found to be inversely correlated with lactic acid, and certain microbial structures were affected by propionic and lactic acid. Overall, there were no correlations between short-chain fatty acids and carbohydrate consumption.

Differential domains and endoproteolytic processing in dominant surface proteins of unknown function from Mycoplasma hyopneumoniae and Mycoplasma flocculare

Summary: Mycoplasma hyopneumoniae possesses surface proteins of unknown function (PUFs) that may play a role in the pathogenicity of porcine enzootic pneumonia (PEP). This study compared the expression of five PUFs in the pathogenic strain 7448 of M. hyopneumoniae with their orthologs from nonpathogenic M. hyopneumoniae J strain and a closely related commensal species Mycoplasma flocculare. Analysis of amino acid sequences and proteomic data revealed differential domains, disordered regions, and repeated motifs. Differential endoproteolytic processing and antigenicity were also observed. Phylogenetic analysis showed higher conservation of three PUFs among Mycoplasma species related to respiratory diseases.
