Summary: Our study suggests that SETDB1 affects microtubule dynamics by interacting with both microtubules and HDAC6 to enhance tubulin deacetylation. These findings provide a novel insight into the cytoplasmic role of SETDB1 in the regulation of microtubule dynamics.

Summary: This study reveals the molecular mechanism of oxygen-dependent degradation of SETDB1 by the CRL2(VHL) E3 complex and its role in genome stability under hypoxic conditions. Loss of SETDB1 leads to the accumulation of transposable element-derived double-stranded RNAs and triggers an immune-inflammatory response and DNA damage-induced cell death.

Summary: SETDB1 regulates lineage-specific genes and endogenous retroviral elements through H3K9me3 deposition. In addition, it co-regulates target gene expression and genome topology with the topological regulator Cohesin at DiSCs, which are regions proximal to gene promoters involved in embryonic stem cell pluripotency and lineage development. This discovery is important for understanding the maintenance and differentiation of embryonic stem cells.

Summary: SETDB1 is essential for bivalent formation and survival of spermatocytes during early meiosis, with its absence leading to aberrant centromere clustering, failure in pairing and synapsis of homologous chromosomes, compromised meiotic silencing of unsynapsed chromatin, and ultimately meiotic arrest and apoptosis of spermatocytes. Additionally, SETDB1 plays a role in repressing transposons and transposon-proximal genes, as well as in regulating meiotic and somatic lineage genes.

Summary: A study revealed the presence of dual regions with both H3K9me3 and H3K36me3 marks in mouse embryonic stem cells, which lose these marks and gain enhancer signatures upon removal of SETDB1, suggesting an important role for heterochromatin in gene control. In differentiated tissues, some of these dual domains become destabilized and enriched in enhancer marks, providing insight into heterochromatin involvement in cell identity maintenance.

Summary: NOP2/NSUN1 catalyzes the deposition of m(5)C at position 4447 on the 28S rRNA, and regulates pre-rRNA processing through non-catalytic complex formation with box C/D snoRNAs. Additionally, NOP2/NSUN1 facilitates the recruitment and stable assembly of U3 and U8 snoRNAs into snoRNP complexes.

Summary: Mutations in the Krebs cycle enzyme IDH1 are associated with better prognosis in gliomas. The study reveals that IDH1 mutant gliomas have diminished expression of the PRMT1-PTX3 axis, leading to dysregulation of autophagy and ferritinophagy. The findings suggest that targeting the PRMT1-PTX3 ferritinophagy regulatory circuit could be a potential therapeutic strategy.

Summary: The SETDB1-TRIM28 complex acts as a critical suppressor of antitumor immunity by negatively regulating PD-L1 expression and activating the cGAS-STING innate immune response pathway to increase infiltration of CD8(+) T cells. Inhibition of SETDB1-TRIM28 bridges innate and adaptive immunity, leading to enhanced antitumor effects of immune checkpoint blockade.

Summary: Genetic and genome-wide analysis of the SETDB1-like enzyme MET-2 in Caenorhabditis elegans reveals its role in promoting transcriptional silencing and fertility through both H3K9 methylation and focus formation. MET-2 has a noncatalytic function that contributes to gene repression. In normal development, MET-2 activity helps maintain fertility, while under heat stress, MET-2 foci disperse, resulting in increased acetylation and transcriptional derepression.
