Summary: A truncated Dicer-like gene CgDCL was identified in oysters, which plays a crucial role in antivirus immunity and can be activated by various stimuli to bind viral nucleic acid. This suggests a potential cooperative relationship between RNAi and IFN-like system in antivirus immunity of oysters.

Summary: This study reveals how a human-blood-derived microRNA, hsa-miR-150-5p, interferes with the mosquito's antiviral system to facilitate flavivirus infection and transmission. By hijacking the mosquito's RNA interference system, this microRNA suppresses the expression of certain chymotrypsins with potent virucidal activity, impacting the transmission of arboviruses.

Summary: The study presents an intracellular delivery approach utilizing cell-penetrating peptide (CPP)-conjugated lipid/polymer hybrid nanovehicles (LPNVs) for ER-targeting, showing efficient uptake by cells and specific ER-targeting ability. Remarkable lysosomal escape of the LPNVs was observed due to effective pH buffering with the aid of PEO-b-PCL-b-PEO, indicating the potential of the LPNVPnt system for elaborate drug delivery technology development at the intracellular level.

Summary: In this study, a near-infrared light-mediated mitochondrial RNA modification approach was developed for long-term imaging and effective tumor suppression. A tumor-targetable NIR fluorescent probe f-CRI was synthesized and it could covalently bind to mtRNA upon 808 nm irradiation, leading to prolonged retention in tumors. The covalent modification of mtRNA by f-CRI disrupted mitochondrial function and significantly suppressed tumor growth. This approach provides a potential strategy for cancer RNA interference therapeutics.

Summary: Gene therapy using tHBcAg VLP as a carrier for shRNA targeting anti-apoptotic Bcl-2 has shown promising results in downregulating Bcl-2 expression in cancer cells. The encapsidated plasmid conjugated with folic acid demonstrated targeted delivery and significant reduction in cell viability, indicating the potential of this approach for gene silencing in cancer therapy.

Summary: This study identified cervical cancer stem cells and revealed the functional changes in cervical stem cells during malignant transformation. The original non-malignant stem cell properties gradually diminished, while the tumor stem cell properties were enhanced. Additionally, stem-like cells in the cervical microenvironment were associated with neoplastic recurrence, and interferon responses were upregulated during lesion progression.

Summary: Cervical cancer is the fourth most common cancer in women globally, with 90% of cancer-related deaths occurring in developing countries. Liquid biopsy may serve as an alternative (or complementary) tool for screening, diagnosis, and follow-up of cervical cancer, including the detection and isolation of circulating tumor cells, cell tumor DNA sequencing, coding and non-coding RNA detection, and exosomal miRNA identification. Further validation and standardization of these methodologies are needed before clinical use.

Summary: Prokaryotes have evolved an immune system, CRISPR-Cas, to defend against invading mobile genetic elements, with different classes and types based on effector Cas proteins and spacer acquisition components. Cas9 and Cas13 are common nucleases used in targeting viral RNA, offering potential applications in virological and biotechnological research.

Summary: This study conducted single-cell RNA and TCR sequencing of cervical cancer tumors and adjacent normal tissues to map the immune landscape. The study revealed tumor-specific germinal center B cells associated with improved clinical outcomes and established a joint model of tumor and stromal cells to predict patients' prognosis. These findings provide information for future combinational immunotherapy.

Summary: This article reviews the systematic processes of sRNA biogenesis, trafficking, and the underlying mechanisms of its functions in plant genomes. sRNAs can move within plants, be transferred to plant pathogens via extracellular vesicles, and be transferred to mammals through food. They function at target sites through various mechanisms.

Summary: Researchers investigated the intracellular trafficking of anti-KRAS antisense oligonucleotides, showing that oligonucleotide AZD4785 is recycled via late endosomes in extracellular vesicles in both cells with poor and good oligo productive uptake. Inducing lysobisphosphatidic acid in late endosomes or blocking EV recycling can enhance AZD4785 activity in cells with poor productive uptake, potentially offering improved treatment strategies.
