Vitamin D receptor is present on the neuronal plasma membrane and is co-localized with amyloid precursor protein, ADAM10 or Nicastrin

Our recent study indicated that vitamin D and its receptors are important parts of the amyloid processing pathway in neurons. Yet the role of vitamin D receptor (VDR) in amyloid pathogenesis is complex and all regulations over the production of amyloid beta cannot be explained solely with the transcriptional regulatory properties of VDR. Given that we hypothesized that VDR might exist on the neuronal plasma membrane in close proximity with amyloid precursor protein (APP) and secretase complexes. The present study primarily focused on the localization of VDR in neurons and its interaction with amyloid pathology-related proteins. The localization of VDR on neuronal membranes and its co-localization with target proteins were investigated with cell surface staining followed by immunofluorescence labelling. The FpClass was used for protein-protein interaction prediction. Our results demonstrated the localization of VDR on the neuronal plasma membrane and the co-localization of VDR and APP or ADAM10 or Nicastrin and limited co-localization of VDR and PS1. E-cadherin interaction with APP or the secretase complex may involve NOTCH1, NUMB, or FHL2, according to FpClass. This suggested complex might also include VDR, which greatly contributes to Ca+2 hemostasis with its ligand vitamin D. Consequently, we suggested that VDR might be a member of this complex also with its own non-genomic action and that it can regulate the APP processing pathway in this way in neurons.

Classical complement pathway factor alterations in narcolepsy

Summary: This study found that the activation of the classical pathway of complement is significant in narcolepsy, suggesting the involvement of immune mechanisms in its pathogenesis. The levels of complement factors and degradation products may serve as biomarkers for predicting the clinical severity of narcolepsy. Further investigation is needed to understand the role of autoantibodies in altering complement levels.

Vitamin D mitigates atherosclerosis risk in TNF-α-induced endothelial cells

Summary: In a cell line model of inflammation, vitamin D was found to decrease the expression of pro-atherosclerotic cytokines and increase the expression of anti-atherosclerotic cytokines, potentially reducing cardiovascular risks. Further verification through in vivo studies is needed.

Maternal and Neonatal Vitamin D Binding Protein Polymorphisms and 25-Hydroxyvitamin D Cutoffs as Determinants of Neonatal Birth Anthropometry

Summary: This study aimed to evaluate the combined effect of maternal and neonatal VDBP polymorphism heterogeneity in conjunction with different maternal and neonatal 25(OH)D cutoffs on neonatal anthropometric profile at birth. The findings suggest a potential role for neonatal VDBP genotypes rs2298850 and rs4588, in conjunction with specific neonatal 25(OH)D cutoffs, in affecting neonatal growth and development. However, no significant effects were observed for maternal VDBP polymorphisms or maternal 25(OH)D concentrations at birth on neonatal anthropometry.

Vitamin D receptor regulates transcription of mitochondrial DNA and directly interacts with mitochondrial DNA and TFAM

Summary: In this study, it was found that Vitamin D receptor (VDR) may function as a mitochondrial transcription factor and regulate the transcription of mitochondrial DNA (mtDNA). The research also revealed the interaction between VDR and mitochondrial transcription factor A (TFAM), suggesting their joint involvement in the regulation of mitochondrial function. These findings not only uncover a novel role of VDR, but also highlight its indispensability for energy-demanding cells.

The relations of circulating agouti-related peptide and leptin with altered sleep architecture in patients with active Cushing's disease: a pilot study

Summary: This study aimed to evaluate the sleep architecture of patients with Cushing's disease (CD) and investigate the role of agouti-related peptide (AgRP) and/or leptin in sleep alterations in patients with active CD. Polysomnography was performed on 26 patients with active CD and compared with age and sex-matched control subjects. The results showed that patients with CD had reduced sleep efficiency and increased wake after sleep onset, and their serum levels of AgRP and leptin were elevated. These findings suggest that CD is associated with impaired sleep efficiency and continuity, which may be influenced by elevated levels of AgRP and leptin.

Chloride intracellular channel protein-1 (CLIC1) antibody in multiple sclerosis patients with predominant optic nerve and spinal cord involvement

CLIC1-antibody may potentially serve as a diagnostic biomarker to differentiate between MS and NMOSD.

Pleiotropic effects of pitavastatin: a pilot study using the saphenous vein endothelial cell model of endothelial injury and prevention of atherosclerosis

Summary: This study compared the efficacy of atorvastatin and pitavastatin in an inflammation model. The results showed that pitavastatin had superior inhibitory effects on certain pro-inflammatory cytokines compared to atorvastatin. This suggests that pitavastatin may be more suitable for controlling cardiovascular diseases caused by atherosclerosis.
