Use of short interfering RNA delivered by cationic liposomes to enable efficient down-regulation of PTPN22 gene in human T lymphocytes

Type 1 diseases based on the selective inhibition of variant PTPN22 allele using lipoplexes of siRNA antisense oligomers.Type 1 diabetes and thyroid disease are T cell-dependent autoimmune endocrinopathies. The standard substitutive administration of the deficient hormones does not halt the autoimmune process; therefore, development of immunotherapies aiming to preserve the residual hormonal cells, is of crucial importance. PTPN22 C1858T mutation encoding for the R620W lymphoid tyrosine phosphatase variant, plays a potential pathophysiological role in autoimmunity. The PTPN22 encoded protein Lyp is a negative regulator of T cell antigen receptor signaling; R620W variant, leading to a gain of function with paradoxical reduced T cell activation, may represent a valid therapeutic target. We aimed to develop novel wild type PTPN22 short interfering RNA duplexes (siRNA) and optimize their delivery into Jurkat T cells and PBMC by using liposomal carriers. Conformational stability, size and polydispersion of siRNA in lipoplexes was measured by CD spectroscopy and DLS. Lipoplexes internalization and toxicity evaluation was assessed by confocal microscopy and flow cytometry analysis. Their effect on Lyp expression was evaluated by means of Western Blot and confocal microscopy. Functional assays through engagement of TCR signaling were established to evaluate biological consequences of down-modulation. Both Jurkat T cells and PBMC were efficiently transfected by stable custom lipoplexes. Jurkat T cell morphology and proliferation was not affected. Lipoplexes incorporation was visualized in CD3+ but also in CD3- peripheral blood immunotypes without signs of toxicity, damage or apoptosis. Efficacy in affecting Lyp protein expression was demonstrated in both transfected Jurkat T cells and PBMC. Moreover, impairment of Lyp inhibitory activity was revealed by increase of IL-2 secretion in culture supernatants of PBMC following anti-CD3/CD28 T cell receptor-driven stimulation. Summary: Autoimmune endocrine disorders such as type 1 diabetes and thyroiditis currently rely on hormone replacement therapy, highlighting the need for personalized immunotherapeutic strategies targeting T and B lymphocytes. This study improved upon a personalized immunotherapeutic approach by functionalizing liposomes with Rituximab antibody, specifically targeting B lymphocytes in conditions like type 1 diabetes. The results showed that these functionalized liposomes had a significant inhibitory effect on the function of B lymphocytes, suggesting potential applicability for personalized immunotherapy in other autoimmune disorders where B lymphocytes are the primary pathogenic immune cells.

Summary: This study investigated the anti-tumor activity of GD2-CAR T-cells in treating DMG/DIPG and identified the potential of using IGF1R/IR inhibitors in combination with GD2-CAR T-cells. The results showed that the dual IGF1R/IR antagonists BMS-754807 and linsitinib were able to inhibit tumor cell viability without affecting CAR T-cells. Linsitinib also improved the function of GD2-CAR T-cells. The combination of linsitinib and GD2-CAR T-cells demonstrated enhanced anti-tumor activity in vitro, ex vivo, and in vivo models of DIPG.

Summary: The development of mixed oxide electrodes is being intensively investigated to reduce cost and improve performance. In this study, thin-film electrodes coated with ruthenium and copper oxide were fabricated. The addition of copper enhanced the capacitive response and electron transfer reversibility, but the interlayer had a negative effect on charge storage power.

Summary: This study focuses on the role of BACH1/NRF2 ratio in the regulation of antioxidant response and its impact on patients with Down syndrome. The results show that overexpression of BACH1 disrupts the induction of antioxidant response genes, leading to oxidative damage accumulation. Additionally, administration of CAPE and VP961 promotes NRF2 nuclear translocation in cells from Down syndrome patients, improving antioxidant response.

Summary: The C1858T variant of the PTPN22 gene is associated with autoimmune diseases. Liposomes carrying siRNA targeting the C1858T variant have been functionalized with a high affinity ligand of Siglec-10, resulting in enhanced delivery to immunocytes expressing Siglec-10. This approach effectively downregulates the variant PTPN22 gene and restores IL-2 secretion in T1D patients.

This study examined the presence of AIRE gene variations in patients with high polyautoimmunity complexity and found a significant association between the S278R polymorphism of the AIRE gene and APECED-like conditions. Additional genetic determinants need to be identified to improve diagnosis and targeted therapies for these patients.

Multifunctional Platform for Covalent Titanium Coatings: Micro-FTIR, XPS, and NEXAFS Characterizations

Summary: This study aims to prepare and characterize a versatile multifunctional platform for immobilizing macromolecules on a titanium surface using robust covalent grafting. Functionalized titanium is widely used in the biomedical field to improve its properties, but long-term stability is compromised due to inflammation and bacterial infections. The proposed method allows for the insertion of three different organic linkers that serve as anchors for biopolymers or bioactive molecules. The use of green solvents in this strategy offers an alternative to traditional solvent methods. Assessment through spectroscopic techniques confirms the successful modification of the titanium surface, and covalent attachment of two polymers shows potential for enhancing biocompatibility and extending the lifespan of biomedical implants.

Summary: A cerium mixed oxide (CeOx) coating was successfully achieved on both pristine and nanotubular-structured Ti and Ti6Al4V surfaces using a simple drop-casting technique. The presence of CeOx on nanotube surfaces enhanced osseointegration, while on non-nanostructured surfaces, this coating improved resistance to oxidative stress and exhibited excellent antibacterial properties.

SARS-CoV-2 infection as possible downstream disease precipitator in autoantibody-positive insulin-dependent diabetes mellitus: a case report
