Reduced Uterine Perfusion Pressure (RUPP) Model of Preeclampsia in Mice

Preeclampsia (PE) is a pregnancy-induced hypertension with proteinuria that typically develops after 20 weeks of gestation. A reduction in uterine blood flow causes placental ischemia and placental release of anti-angiogenic factors such as sFlt-1 followed by PE. Although the reduced uterine perfusion pressure (RUPP) model is widely used in rats, investigating the role of genes on PE using genetically engineered animals has been problematic because it has been difficult to make a useful RUPP model in mice. To establish a RUPP model of PE in mice, we bilaterally ligated ovarian vessels distal to ovarian branches, uterine vessels, or both in ICR-strain mice at 14.5 days post coitum (dpc). Consequently, these mice had elevated BP, increased urinary albumin excretion, severe endotheliosis, and mesangial expansion. They also had an increased incidence of miscarriage and premature delivery. Embryonic weight at 18.5 dpc was significantly lower than that in sham mice. The closer to the ligation site the embryos were, the higher the resorption rate and the lower the embryonic weight. The phenotype was more severe in the order of ligation at the ovarian vessels < uterine vessels < both. Unlike the RUPP models described in the literature, this model did not constrict the abdominal aorta, which allowed BP to be measured with a tail cuff. This novel RUPP model in mice should be useful for investigating the pathogenesis of PE in genetically engineered mice and for evaluating new therapies for PE.

Factors Associated with the Antihypertensive Effect of Esaxerenone and Serum Potassium Elevation: A Pooled Analysis of Seven Phase III Studies

Summary: This study examined factors associated with the antihypertensive effects of esaxerenone and the incidence of serum potassium elevation in patients with hypertension. Results showed that factors such as sex, weight, hypertension duration, prior antihypertensive treatment, plasma aldosterone concentration, urinary albumin-to-creatinine ratio, and smoking were associated with changes in blood pressure. Factors significantly associated with serum potassium elevation included baseline serum potassium, renal function, urinary albumin-to-creatinine ratio, blood pressure, and hypertension grade. The study concluded that esaxerenone is effective for patients with various backgrounds, but careful management is needed for those at risk of elevated serum potassium.

Summary: Unlike mammals, avian early embryos have PGCs that utilize blood circulation to migrate to the somatic gonadal primordium. Our research shows that PGCs are arrested at a specific site in the capillary plexus, enabled by increased stiffness mediated by actin polymerization. They then soften to squeeze through the endothelial lining.

Region-based SVD processing of high-frequency ultrafast ultrasound to visualize cutaneous vascular networks

Summary: Observing alterations in cutaneous vasculature can be a potential diagnostic marker for diseases. High-frequency ultrasound (HFUS) has been used in dermatology to observe skin morphologies and tissue conditions, but its ability to visualize micro-vessels is limited. In this study, a novel region-based SVD filtering approach was proposed to visualize cutaneous vascular networks. Experimental observations showed that this approach improved the image quality and has potential applications in the diagnosis of cutaneous disorders.

Development of Ultrasound Phantom Made of Transparent Material: Feasibility of Optical Particle Image Velocimetry

Summary: A transparent ultrasound flow phantom material composed of poly(vinyl alcohol) hydrogel (PVA-H) with dimethyl sulfoxide (DMSO) and water solution was proposed for visualizing and measuring flow inside ultrasound systems. The material's refractive index was modified to match that of glass for transparency. Feasibility tests were conducted, and an ultrasound flow phantom was fabricated for B-mode visualization and Doppler-PIV comparison. The results showed promising accuracy in velocity measurement and similarity to real tissue visualization, with some limitations regarding sound velocity and Doppler measurements.

Microfluidic platform for the reproduction of hypoxic vascular microenvironments

Summary: This study developed a microfluidic platform for reconstructing hypoxic vascular microenvironments, by simultaneously applying hypoxic stress and fluid shear stress. The results showed that under the simultaneous exposure to hypoxic stress and fluid shear stress, the migration velocity of endothelial cells increased and gradually decreased, reaching the lowest value. Meanwhile, the cells aligned and elongated in the flow direction, with enhanced expression of VE-cadherin and actin filament assembly. Therefore, this microfluidic platform is useful for investigating the dynamics of endothelial cells in vascular microenvironments.

Numerical analysis of hemodynamic changes and blood stagnation in the left ventricle by internal structures and torsional motion

Summary: This study investigated the effects of complex internal structures and torsional motion on left ventricular hemodynamics using computational fluid dynamics analyses. The presence of trabeculae carneae and papillary muscles decreased average wall shear stress and increased the relative residence time of blood stagnation at the apex of the left ventricle. Torsional motion caused irregular changes to the relative residence time level and distribution at the apex. Therefore, the complex internal structures and torsional motion of the left ventricle could cause blood stagnation.

A Review of Functional Analysis of Endothelial Cells in Flow Chambers

Summary: Vascular endothelial cells are exposed to mechanical stress from blood flow and their functions can be studied using specific chamber designs. The functions of these cells are closely connected to the outer layers of arteries and organs, and the interaction between smooth muscle cells in the middle layer and endothelial cells can be investigated using coculture chambers. Additionally, microfabrication technology in microfluidic devices allows for mechanical and chemical stimulation of cells to study their dynamics in vivo microenvironments.
