Summary: GLP-1 receptor agonists (GLP-1RAs) have been developed as therapeutic agents for type 2 diabetes (T2D) since 2005. GLP-1R is expressed not only in pancreatic islets but also in other organs, especially the lung. The controversy surrounding extra-pancreatic GLP-1R expression still needs to be resolved, and more reliable GLP-1R antibodies should be utilized for further research. The anti-inflammatory features of GLP-1RAs have shown promising results, but whether these features are directly mediated by GLP-1R expressed in immune cells remains controversial.

Summary: Glucotoxicity, impaired insulin secretion, suppression of insulin gene expression, and apoptosis in pancreatic beta-cells caused by chronic hyperglycemia are key factors in the development of type 2 diabetes. Rare sugar d-allulose has been found to have antihyperglycemic and antihyperlipidemic effects. However, it is unclear how rare sugars, including d-allulose, directly affect pancreatic beta-cell function. This study shows that chronic exposure to certain ketohexoses can lead to reduced insulin gene expression, but d-allulose, d-sorbose, l-fructose, and l-tagatose do not affect pancreatic beta-cell function.

Summary: This study identified a novel small molecule GLP-1R agonist, S6, through virtual screening and calcium analysis techniques. Data from cellular and biophysical experiments indicated that S6 effectively bound to GLP-1R, increased intracellular calcium concentration by GLP-1R activation, inhibited Kv channels, and stimulated glucose-dependent insulin secretion. These findings provide valuable insights for future development of small-molecule agents targeting GLP-1R.

Summary: This study aimed to determine the kinase activity profiles of human pancreatic beta cells downstream of glucagon-like peptide-1 receptor (GLP-1R) balanced versus biased agonist stimulations. The results showed that acute and sustained GLP-1R agonist exposure had distinct effects on kinase responses, and different biased agonists could modulate distinct kinase interaction networks. These findings have important implications for the selection of appropriate anti-type 2 diabetes therapies.
