Structure of Utp21 Tandem WD Domain Provides Insight into the Organization of the UTPB Complex Involved in Ribosome Synthesis

Assembly of the eukaryotic ribosome requires a large number of trans-acting proteins and small nucleolar RNAs that transiently associate with the precursor rRNA to facilitate its modification, processing and binding with ribosomal proteins. UTPB is a large evolutionarily conserved complex in the 90S small subunit processome that mediates early processing of 18S rRNA. UTPB consists of six proteins Utp1/Pwp1, Utp6, Utp12/Dip2, Utp13, Utp18 and Utp21 and has abundant WD domains. Here, we determined the crystal structure of the tandem WD domain of yeast Utp21 at 2.1 A resolution, revealing two open-clamshell- shaped beta-propellers. The bottom faces of both WD domains harbor several conserved patches that potentially function as molecular binding sites. We show that residues 100-190 of Utp18 bind to the tandem WD domain of Utp21. Structural mapping of previous crosslinking data shows that the WD domains of Utp18 and Utp1 are organized on two opposite sides of the Utp21 WD domains. This study reports the first structure of a UTPB component and provides insight into the structural organization of the UTPB complex.

Crystal structures and insights into precursor tRNA 5'-end processing by prokaryotic minimal protein-only RNase P

Summary: In this study, the crystal structure of HARP proteins and their mechanism of pre-tRNA binding and cleavage were elucidated. The results showed that HARP proteins can undergo conformational changes to facilitate pre-tRNA binding and catalytic site formation, and can assemble into larger oligomers. The study also revealed a cooperative pre-tRNA processing mode, where one HARP dimer binds pre-tRNA at the elbow region and 5'-end removal is catalyzed by the partner dimer.

Reconstitution of Multi-Protein Complexes through Ribozyme-Assisted Polycistronic Co-Expression

Summary: A Ribozyme-Assisted Polycistronic co-expression system (pRAP) was developed for heterologous co-production and in vivo assembly of multi-subunit complexes. Summary: The human AAA+ ATPase CLPB is a protein disaggregase in the mitochondrial intermembrane space and plays a role in promoting the solubilization of mitochondrial proteins. Structural characterization reveals that CLPB forms different oligomers depending on its state and contains an ANK domain essential for its disaggregase activity. Interactome analysis suggests that the ANK domain may directly interact with various mitochondrial substrates. These findings highlight the unique properties of CLPB and its potential as a therapeutic target for mitochondria-related diseases.

Structural basis of a two-step tRNA recognition mechanism for plastid glycyl-tRNA synthetase

Summary: Two types of glycyl-tRNA synthetase (GlyRS) have been identified, alpha(2) and alpha(2)beta(2) GlyRSs. While the mechanisms for alpha(2) GlyRSs are well understood, little is known about the mechanisms for alpha(2)beta(2)/(alpha beta)(2) GlyRSs. This study presents the structures of the (alpha beta)(2) GlyRS from Oryza sativa chloroplast, revealing a complex mechanism of tRNA selection and access to the active site.
