Summary: This study investigated the epigenetic-mediated mechanism of the PAX1 gene in sporadic parathyroid adenomas. The results revealed that the expression of PAX1 was significantly reduced in parathyroid adenomas, which was associated with hypermethylation in the promoter region and lower levels of histone modification. Treatment with a pharmacologic inhibitor led to the re-expression of the PAX1 gene. These findings provide insights into the epigenetic deregulation of PAX1 in parathyroid adenomas and have potential clinical and therapeutic implications for patients with PHPT.

Summary: METTL14 promotes global genome repair and suppresses UVB-induced skin tumorigenesis by regulating DDB2 translation through m6A mRNA methylation, and its downregulation by UVB irradiation through NBR1-dependent selective autophagy plays a critical role in skin cancer development.

Summary: Polyamines, such as spermidine and spermine, have physiological activities similar to antioxidants and anti-inflammatory substances. Increased intake of polyamines can inhibit aging-associated pathologies and extend lifespan. The relationship between polyamine metabolism and DNA methylation plays a role in regulating gene expression and promoting healthy longevity.

Summary: The m(6)A reader YTHDC2 is frequently suppressed in lung adenocarcinoma, leading to poor clinical outcomes, and it exerts antitumor activity by suppressing cystine uptake via system XE. Downregulation of YTHDC2 destabilizes SLC7A11 mRNA in an m(6)A-dependent manner, implicating the modulation of cystine uptake in promoting lung adenocarcinoma tumorigenesis.

Summary: This study found that SNORA23 regulates ribosome biogenesis in hepatocellular carcinoma (HCC) by impairing the 2'-O-ribose methylation of 28S rRNA. SNORA23 inhibits the proliferation, migration, and invasion of HCC cells in vitro and in vivo. It is regulated by the PI3K/Akt/mTOR signaling pathway and inhibits the phosphorylation of 4E binding protein 1.

Summary: This study describes a subset of glioblastoma characterized by hypomethylation at specific regions, suggesting a shared ontogeny between these tumors and astroglial progenitors. The presence of astrocytic gene signature and predicted binding sites for transcription factors involved in demethylation and astroglial lineage specification further support this finding. Functionally, this subset of glioblastoma is associated with increased invasiveness mediated by SRPX2 and macrophage infiltration.

Summary: The study reveals that TBX20 is downregulated in early-stage colorectal carcinoma (CRC) tissues and is associated with poor prognosis. TBX20 suppresses cell proliferation and tumor growth in CRC cells by inhibiting the non-homologous DNA end joining (NHEJ)-mediated repair. This research uncovers the tumor-suppressive role of TBX20 in CRC and provides a potential biomarker for predicting prognosis and a therapeutic target for combination therapy in early-stage CRC patients.

Summary: The study reveals a novel role of human EAF1 in regulating transient transcriptional inhibition during genotoxic stress, by enhancing ELL self-association and reducing its interaction with other Super Elongation Complex (SEC) components. This mechanism describes an important dynamic transcriptional regulation involving post-translational modification of a key elongation factor.

Summary: The study showed that SETD6 is significantly upregulated in oral squamous cell carcinoma (OSCC) tissues. Silencing SETD6 inhibited proliferation, migration, and invasion of OSCC cells, induced apoptosis, and resulted in G1 arrest. Additionally, SETD6 siRNA also suppressed promoter methylation of RelA and PAK4.

Summary: Chronic pain increases the risk of anxiety, with limbic areas likely being involved. This study investigates the behavioral, hormonal, and neuroplastic correlates of anxiety in persistent pain. The results show that persistent inflammatory pain promotes anxiety-like behaviors, HPA axis activation, and epigenetic regulation through DNA methylation in the amygdala.

Summary: This study compared the expression profile of DNA methyltransferases and the global DNA methylation status in different phases of multiple myeloma (MM) patients. The expression of DNMT1 was unchanged, while DNMT3A was overexpressed in both sorted and unsorted cell populations of MM patients. The global DNA methylation status did not correlate with the clinical stage of MM in unsorted samples. This study provides insights into the potential role of DNMT3A as a marker for MM tumor progression.
