Summary: N-myc downstream-regulated gene 2 (NDRG2) is a tumor-suppressor gene that inhibits tumorigenesis and metastasis of tumors. Loss of NDRG2 expression is associated with higher tumor grade, increased metastasis, and decreased patient survival. NDRG2 inhibits multiple cell signaling pathways involved in tumor metastasis and activates GSK-3 beta, which has anti-tumor effects. Although the molecular mechanism of NDRG2 as an adaptor protein is not fully understood, it holds potential as a molecular target for inhibiting tumor metastasis and improving patient prognosis.

Summary: Piceamycin (1), a macrocyclic lactam isolated from the silkworm's gut, has been found to possess antibacterial activity. This study investigates the potential anticancer activity of 1 and its underlying mechanisms in colorectal cancer (CRC) cells. The results show that 1 inhibits CRC cell growth by upregulating NDRG1, inducing cell cycle arrest and apoptosis, and inhibiting cell migration. The disruption of the interaction between NDRG1 and c-Myc is also observed. This suggests that 1 may serve as a potential candidate for CRC treatment by regulating NDRG1-mediated functionality.

Summary: POM121 is overexpressed in gastric cancer tissues compared to normal gastric tissues and is associated with deep invasion, distant metastases, and TNM stage in gastric cancer patients. POM121 promotes proliferation, migration, and invasion of gastric cancer cells through the phosphorylation of the PI3K/AKT pathway and increased expression of MYC. These findings suggest that POM121 may serve as an independent prognostic factor for gastric cancer patients.
