Summary: This article reviews the regulation of RTKs/TGF beta R between PDAC and CAFs, as well as the RTKs/TGF beta R inhibitor-based clinical trials on pancreatic cancer. It discusses the importance of considering the signaling of receptor tyrosine kinases and TGF beta receptors in overcoming the obstruction in the tumor microenvironment. The impact of receptor activation on cell cycle regulation, signal transduction pathways, and apoptotic sensitivity of tumor cells is also highlighted.

Summary: TGF-beta plays essential roles in early embryonic development, organogenesis, immune regulation, tissue repair, and adult homeostasis. Its involvement in fibrosis and cancer is complex and context-dependent, showing both inhibitory and promoting effects. Therapeutics targeting TGF-beta signaling hold promise in treating fibrosis and cancer, although the development is hindered by potential systemic cytotoxicity.

Summary: This study revealed a novel mechanism of integrin beta 1 in tumor growth by modulating the expression of kindlin-2 and TGF-beta receptor 2 signaling. Loss of integrin beta 1 affects cell proliferation and adhesion in a 3D matrix, thus impacting tumor growth.

Summary: This study found that the population of M2-TAMs and VEGFR2 expression significantly increased in the tumors of CRC patients. HIF1-alpha-positive cells were significantly correlated with the VEGFR2 expression level on M2-TAMs. In vitro experiments demonstrated that M2 macrophages produced TGF-beta 1 through the VEGF/VEGFR2 signaling pathway.

Summary: In this study, we identified dihydroergotamine (DHE) as a potential drug for treating liver fibrosis by targeting TGF beta R2 and inhibiting the TGF beta signaling pathway. DHE was shown to reduce fibrotic gene expressions and improve liver fibrosis in both cellular and mouse models.

Summary: In this study, the anti-fibrotic effects of derrone on TGF-beta 1-stimulated MRC-5 lung fibroblast cells and bleomycin-induced lung fibrosis were investigated. It was found that high concentrations of derrone increased cytotoxicity in MRC-5 cells, but low concentrations did not induce significant cell death. Derrone reduced the expression of TGF-beta 1, fibronectin, elastin, and collagen1 alpha 1 in TGF-beta 1-stimulated MRC-5 cells, and also attenuated fibrotic histopathological changes in bleomycin-treated mice.

Summary: Diabetes is caused by insufficient insulin production or loss of β-cells. Restoring regulated insulin production is the main goal of translational diabetes research. Recent advances include β-cell proliferation, regeneration, and replacement, as well as the discovery of therapeutic targets and small molecules. Promising strategies have been found in β-cell transdifferentiation, neogenesis, and stem cell differentiation. Clinical trials using in vitro-generated β-like cells are underway. Continued support is critical for the development of distinct diabetes therapies.

Summary: This pilot study explored the potential role of drain fluid cytokines in predicting postoperative pancreatic fistula (POPF) after pancreaticoduodenectomy. The results showed that the levels of serum amylase, drain fluid amylase, TGF-beta, and EGF on postoperative day 1 were significantly higher in patients developing POPF. Assessing the local inflammatory response after pancreatoduodenectomy could be a promising field of research.
